| Objective:To investigate the targeting property and correlative factors of the FITC-CSNRDARR C molecular probe labeling orthotopic transplantation tumor of bladder cancer in vivo.Methods:From July 2013 to June 2014,the stability and absorption spectrum of FITC-CSNRD ARRC molecular probe were detected by spectrophotometer and molecular imaging to det ermine the optimum concentration and imaging time window. 30 BALB-C nude mice wer e randomly divided into experimental group(n=20)and control group(n=10).In control gro up,5 of them(group A) were ligated ureter, others(B group) were not. We established orth otopic transplantation tumor model in surgical operation, and then injected 0.2m L probes(220μmol/L) intravenously after 2 weeks. We obtained images and analyzed average gray va lue of the organs and orthotopic transplantation tumor by using optical probe molecule fluo rescence imaging system after 30 min,1h,2h,4h and 12 h respectively.Results:After injected the FITC-CSNRDARRC molecular probes intravenously at 220μmol/L, the fluorescence signal of tumor tissue strengthened gradually.The intensity of fluoresc ence signal was strongest at 4h. The illumination and temperature had little effect on the fl uorescence signal. The average gray value of tumor tissue at 1hã€2hã€3hã€4hã€5hã€6hã€8hã€12h were 74.22ã€76.2ã€80.11ã€89.38ã€83.29ã€85.1ã€81.22ã€83.01, respectively. Th e fluorescence signal of normal tissue weakened gradually with the passage of time.Conclusions:The fluorescence speciality of the probe is affected by concentration, and the optimu m concentration of labeling tumor is 220μmol/L. The optical imaging time window was ab out 4 h after intravenous injection. The FITC-CSNRDARRC molecular probe can specifica lly bound to orthotopic transplantation tumor of bladder cancer in vivo. |
