Objective:Study on bladder tumor homing peptide NYZL1-FITC optical molecular probe for bladder perfusion,stability in urine,saline,Mannitol Injection,blood,and temperature,p H,light(white,Lv Guang)influence on NYZL1-FITC optical molecular probe of the molecular structure,it is proposed that NYZL1-FITC optical molecule probe is intraperitoneally instilled after intraperitoneal injection of specific molecular markers,and to provide a theoretical basis for improving the recognition rate of small tumor.Methods:The concentration of the optical molecular probe(μmol / L)in the bladder tumor targetting peptide-FITC optical molecular probe solution was measured by high performance liquid chromatography(HPLC)in order to understand the urine,blood,saline,3% The effects of mannitol injection,light(white,green),temperature and p H on the stability of bladder tumor targetting peptide-FITC.Each experiment was repeated three times.Results:(1)NYZL1-FITC solution was incubated with urine for different time(0min,15 min,30min,60 min,120min),the concentration of NYZL1-FITC was 175.0±1.0,173.3±1.5,172.0±1.7,171.7±1.5,170.3±1.5 respectively by high performance liquid chromatography,the single factor analysis of variance of repeated measurement,NYZL1-FITC concentration had no significant change in 120min(F= 6.91,P >0.01).(2)NYZL1-FITC solution was incubated with serum for different time(0min,15 min,30min,60 min,120min),the concentration of NYZL1-FITC was 175.0±1.0,76.0±2.0,72.7 ± 1.5,68.7 ± 1.5,66.0 ± 1.0 respectively by high performance liquid chromatography.The variance analysis was performed by single factor and repeated analysis.The concentration of NYZL1-FITC changed significantly within120min(F= 3072.19,P<0.01).(3)NYZL1-FITC solution was incubated with normal saline for different time(0min,15 min,30min,60 min,120min),the concentration of NYZL1-FITC was 175.0±1.0,172.0 ± 1.0,170.0 ± 1.0,169.3 ± 1.5,169.0 ± 1.0respectively by high performance liquid chromatography.The variance analysis was performed by single factor and repeated analysis.The concentration of NYZL1-FITC changed significantly within 120min(F= 14.66,P<0.01).(4)NYZL1-FITC solution was incubated with3% Mannitol Injection for different time(0min,15 min,30min,60 min,120min),the concentration of NYZL1-FITC was175.0±1.0,174.3±0.6,173.0±1.0,173.0±1.0,171.0±1.2 respectively by high performance liquid chromatography.The variance analysis was performed by single factor and repeated analysis.The concentration of NYZL1-FITC had no significant change in 120min(F= 6.91,P > 0.01).(5)NYZL1-FITC solution was incubated at 25 ℃(room temperature),30 ℃,37 ℃,40 ℃ for 30 min,the concentration of NYZL1-FITC was 350.0 ± 1.0,349.0 ±1.0,349.7 ± 1.5,348.7 ± 0.6 respectively by high performance liquid chromatography.The variance analysis was performed by single factor and repeated analysis.The concentration of NYZL1-FITC had no significant change in120min(F= 0.952,P > 0.01).(6)NYZL1-FITC solution was incubated by white light 0min,1min,5min,10 min,the concentration of NYZL1-FITC was 350.0±1.0,260.3±2.5,65.0±3.0,26.0±1.0respectively by high performance liquid chromatography.The variance analysis was performed by single factor and repeated analysis.The concentration of NYZL1-FITC changed significantly within 120min(F= 9548.52,P<0.01).(7)NYZL1-FITC solution was incubated by green light 0min,1min,5min,10 min,the concentration of NYZL1-FITC was 350.0±1.0,260.3±2.5,65.0±3.0,26.0±1.0respectively by high performance liquid chromatography.The variance analysis was performed by single factor and repeated analysis.The concentration of NYZL1-FITC changed significantly within 120min(F= 16663.23,P<0.01).(8)NYZL1-FITC solution was incubated at PH value of 5,6,7,8,9 for 30 min,the concentration of NYZL1-FITC was 173.0±1.0,172.7±1.5,175.0 ±1.0,173.0±2.0,170.0 ± 1.0 respectively by high performance liquid chromatography.The variance analysis was performed by single factor and repeated analysis.The concentration of NYZL1-FITC had no significant change in 120min(F=5.13,P >0.01).Conclusion:The physicochemical properties of NYZL1-FITC optical molecular probe,at 25-40 DEG C and p H 9 5-urine,normal saline,3% Mannitol Injection incubated with no change in 120 min,but the effective concentration in serum gradually decreased,White light and wavelength of 535 nm green light for 10 min,NYZL1-FITC optical molecular probe effective concentration decreased rapidly,the fluorescence intensity decreased,indicating that long time light will affect the NYZL1-FITC optical molecular probe physical and chemical properties,the mechanism to be further studied.
