Purposes Study the expression of female hormonal receptors:estrogen receptors(ER),progesterone receptor(Pg R) and transforming growth factor- beta 1(TGF-β1) in breast fibroadenoma tissue and normal tissue around, exploring their role in the pathogenesis of breast fibroadenoma.Methods 447 pairs paraffin slids were from the archived samples after mammotome or resection operations and were pathological diagnosed with breast fibroadenomas. The expression of ER and Pg R were analysed with immunohistochemica L(IHC) S-P method, //’sample includes tumor tissue and normal tissue around. Another 20 pairs fresh tissue specimens( each included tumor tissue and normal tissue around) were collected from the patients with informed consent and were preserved through liquid nitrogen, all these samples were used reverse transcription quantitative polymerase chain reaction(RT-q PCR) to detect TGF-β1 m RNA level after being diagnosed of fibroadenoma. All the IHC results were analysed with SPSS 16.0 statistical software. ER, Pg R expression differences between the two groups using nonparametric Wilcoxon test inspection level(P<0.05); The correlation coefficient between age and hormonal receptors expression adopted Spearman correlation analysis, inspection level(P<0.05). The TGF-β1 m RNA level analysis were used variance analysis, P<0.05 for the statistical difference. Results The ER/Pg R expression of tumor group was obviously higher than that of normal tissue, The differences were statistically significant. The correlation coefficient of ER/Pg R expression between tumor tissues and age, no statistical significance. The correlation coefficient of ER expression in normal tissue and age is a statistical significance(P < 0.05),with age increased, the ER expression decreased. Although a trend was observed that with the age increasing,the Pg R expression in normal tissue decreased, but there was no statistical significance. The RT-q PCR results show breast fibroadenoma tissue TGF-β1 m RNA expression level is higher than the tumor adjacent tissues(P=0.03).Conclusion Female breast fibroadenoma glandular epithelium average hormone receptors expression was significantly higher than that of the surrounding normal tissues, suggests that the high expression of hormone receptors in breast fibroadenoma, plays an important role in the pathogenesis and breast fibroadenoma tissue. Fibroadenoma hormone receptors, unlike normal tissue, with the increased age, the expression less, show that the high hormone receptors expression is autonomy. TGF-β1 m RNA expression level was higher than the peritumoral tissue, shows that TGF-β1 as an important factor for the development of stroma also involved in the formation process of fibroadenoma. The exact process remains to be further researched.
