Objective:To investigate the effects of sevoflurane preconditioning combined with postconditioning on expression of angiotensin converting enzyme(ACE)m RNA in lung tissues during lung ischemia-reperfusion. Investigate the protective effect on lung ischemia-reperfusion injury in sevoflurane preconditioning combined with postconditioning.Methods:Choose good health, clean grade of Xinjiang Medical University Experimental Animal Center of,SD rats weighing between 250 ~ 350 g, calculated in this study required a total of 54 rats in each group 18.The rats were randomly according randomly divided into three groups(n = 18). Sham group(S group): rats were anesthetized by intraperitoneal injection back in the prone position, the free rat trachea, endotracheal intubation, then small animal ventilator mechanical ventilation for 30 min, rats were on the right lateral position, and in the fifth intercostal rat left chest, left lung door free, and application preparation S group lung tissue exposed wet gauze soaked in 0.9% sodium chloride injection ever; lung ischemia-reperfusion group(I / R group): S group using the same method for preparing the same model model rat lungs were exposed to rats expiratory noninvasive blood vessels free of the rat out of the left lung door(including the left main bronchus, left pulmonary artery and vein) were clipped. In rats hilar blocked after 45 min, loosen lung tissue forceps restore blood circulation and ventilation, formation of lung ischemia-reperfusion model;sevoflurane preconditioning combined after-treatment group(SPr + o group): In front of the door to block the front of the left lung of rats inhaled 2.1% sevoflurane 30 min, eluting 10 min, then blocked the left lung door 45 min, and in the formation of reperfusion immediately sucked again by 2.1% heptafluorobutyric alkyl 30 min, this model sevoflurane combined treatment group was prepared. When we were sacrificed and reperfusion respectively 30, 60 and 120 min 6 rats, the lung tissue of rats was measured in rat lung tissue wet weight / dry weight ratio(W / D ratio) of hematoxylin- eosin France(hematoxylin-eosin staining, HE)histological examination, colorimetry MDA(malondialdehyde, MDA) expression levels, ELISA(enzyme-linked immunosorbent assay, Elisa) plasma endothelin-1( endothelin-1, ET-1) expression levels,the expression ACEm RNA measured by RT-PCR.Results:Compared with the S group, Spr+o group and I /R group at each time point reperfusion lung tissue W / D, ACEm RNA expression and plasma ET-1, MDA levels were increased(P <0.05). Compared with the I / R group, Spr + o group at each time point reperfusion lung tissue W / D ratio, ACEm RNA expression and plasma ET-1, MDA levels were decreased(P <0.05). Microscopic examination showed that the pathologic changes were significantly attenuated in group Sr+o compared with group I/RConclusion : The mechanism of sevoflurane preconditioning combined with postconditioning for lung ischemia-reperfusion injury protection may be down ACEm RNA, ET-1 expression and reduce oxygen free radicals.
