BackgroundHand-foot-mouth disease is a kind of class C infectious diseases caused by enterovirus, and it often affects children under 5 years of age, especially under 3 years of age. The disease can spread all over the world, and occur throughout the year, more in summer and autumn. This disease has become one of the most important infectious diseases affecting children’s physical and mental health in the world.ObjectiveHand-foot-mouth disease patients’ stool specimens in 2010-2014 and healthy people’stool specimens in 2012 were detected using RT-PCR and Real Time RT-PCR, and partial stool specimens in 2013-2014 were detected using cell culture isolation technique. Detective results showed that there was a positive correlation between the latent infection rate and enterovirus epidemic type. And these detective results provided a reliable scientific basis for developing hand-food-mouth disease’prevention and control strategy of Linyi.MethodsHand-foot-mouth disease patients’ stool specimens in 2010-2014 were detected using RT-PCR and Real Time RT-PCR, and partial stool specimens in 2013-2014 were detected using cell culture isolation technique, and healthy people’ stool specimens from scattered and kindergartens’children under 5 years of age and guardian of children under 3 years of age in 2012 were detected using Real Time RT-PCR.Main resultsIn the paper, hand-foot-mouth disease patients’stool specimens in 2010-2014 and healthy people’stool specimens in 2012 were detected using RT-PCR, Real Time RT-PCR and ell culture isolation technique. Many results were obtained through analyzing detective results, as follows.1. Enterovirus 71 and coxsackievirus A16 had alternately become epidemic strain in Linyi city from 2010 to 2014. And this had basically become a epidemic regularity of hand-food-mouth disease in Linyi city.2.123 of 1275 healthy people’ stool specimens were positive PE and CA16, and positive rate was 9.65%, and 111 specimens were positive PE, and positive rate was 8.71%,12 specimens were positive CA16, and positive rate was 0.94%, and there was not one positive CA16 specimens.3. Enteroviral latent infection rate from 6 of 7 age groups’research objects of Lanshan district was higher than Pingyi county, apart from 5-6 year old children, especially 4-5 year old children and children under 1 year of age.4. Enteroviral latent infection rate of healthy people of Lanshan district were higher than Pingyi county in May, July and October. There was a bell of change in proportion of the enteroviral latent infection following season in Lanshan and Pingyi county.5. There was a positive correlation between the enteroviral latent infection rate of PE and CA16 and entero virus epidemic type.Conclusion and adviceThe paper gained three conclusions through analyzing hand-foot-mouth disease patients’and healthy people’stool specimens PCR detective results in 2010-2014, as follows. Firstly, entero virus 71 and coxsackievirus A16 had alternately become epidemic strain in Linyi city from 2010 to 2014; Secondly, there was a bell of change in proportion of the enteroviral latent infection following season in Linyi city; Finally, there was a positive correlation between the enteroviral latent infection rate and entero virus epidemic type in Linyi city.A series of measures were performed to prevent epidemic when enterovirus 71 became epidemic. Firstly, the high attention of leaders should be won over, and a responsibility for prevention and treatment of hand, foot and mouth disease was made clear; Secondly, preventive and control measures of hand, foot and mouth disease were carried out, especially, kindergartens; Thirdly, hospital should carry out medical rescue work actively and earnestly; Finally, kindergarten and primary school health education should be strengthened, and parents of disease prevention awareness should be improved.
