Objective:Observation of the change of liver function and serum fibrosis index to validate the curative effect of touxietongluo from the Experimental Research,Methods:104 male Wistar rats were randomly divided into four groups: blank model group (24),model group (32), touxietongluo group (24,treated by modified sanjia powder decoction), positive control group (24, treated by fufangbiejiaruanganpian). Except blank model group, the other 3 groups conducted by intraperitoneal injection of pig serum to estblish immune liver fibrosis model.8 weeks after model establishment, the rats in the blank model group and model group were given saline by gavage, the rats in the touxietongluo group were given modified sanjia powder decoction by gavage, the rats in the positive control group were given fufangbiejiaruanganpian by gavage.6 rats in each group were drawn blood from the heart, and take the liver tissues after 30,60, and 90 days after the treatment. ELISA was used to detected serum liver function and liver fibrosis index. The pathological morphological changes of liver tissues were observed HE stain and electron microscopy.Results:1 General healthy condition The rats of blank model group was active with shining fur, drank and ate normally, weight increased regularly. The rats of model group was weary with dim fur, weight increased slowly, or lose weight during the experiment, and some of them died of weakness at the later stage of the experiment. Before the treatment, the physical condition of the touxietongluo group and positive control group was as same as the rats of model group at the same stage of the experiment, after the treatment the rats became healthier, the fur became shining, weight increasing with the treatment time went on, but comparing them with the rats of blank model group the weight still less.2. Pathomorphological changesThe livers of the rats in the model group was darker on the surface with small nodules, size did not change obviously. Comparing with blank model group, the livers had a thicker edges, close texture, parts of the liver had adhesions with omentum majus. Observed under light microscope,itshowed that the normal liver tissue had been damaged, mainly expressed cell degeneration and necrosis, inflammatory cell infiltration, Hepatocyte swelling, steatosis hepatocyte, and mass of proliferation of fibrous tissue made a disordered structure of hepatic lobule, fale liver acinus trend forming. Lymphocytic and monocyte infiltration were found in fibrous septa.The liver function index and fibrosis index of the rats in control group did not have obvious abnormality.After 60 days of treatment, compared to the model group the HA, LN, Pâ…¢NP of the touxietongluo group and positive control group were significantly decreased while the AST showed a significant difference (P<0.05).And those difference till existed after 90 days of treatment. Other detection index had no significant difference (P>0.05). Also,there was no significant difference between touxietongluo group and positive control group (P>0.05).Conclusions:1. Immune complex-induced animal model of liver fibrosis can be established by intraperitoneal injection of pig serum.2. Modified Sanjia Powder decoction could improve the liver function,fibrosis index, and pathological of liver.
