Objective: Previous studies have confirmed that the biansetong mixture is effective in the treatment of chronic constipation, the mechanism of action is not clear. The aim of this study was to explore the regulating effects and mechanisms of BST serum containing on the expression of AQP3 in cultured HT-29 cells in vitro.Methods: 36 adult healthy SD rats, sex in half, were randomly divided into ultra pure water group,low-dose,middle-dose and high-dose groups of BST.The low-dose,middle-dose and high-dose groups of BST were used as the 1.67g/kg,23.34g/kg and 35.00g/kg. The rats were anesthetized after 7 days of intragastric administration and blood samples were obtained from abdominal aorta to prepare the BST serum containing. HT-29 cells cultured in vitro were treated with different concentrations of BST serum containing for24 hours. The expression levels of protein and mRNA of AQP3 were decided by Western Blot and reverse transcription-polymerase chain reaction.Results: The low-dose,middle-dose and high-dose groups of BST serum containing can reduce HT-29 cells AQP3 protein and mRNA expression level(P < 0.01). The ability of middle-dose group of BST serum containing to down regulate HT-29 cells AQP3 mRNA and protein expression level is stronger than that of low-dose groups of BST serum containing(P < 0.01).The ability of high-dose group of BST serum containing to down regulate HT-29 cells AQP3 mRNA and protein expression level is stronger than that oflow-dose and middle-dose groups of BST serum containing(P < 0.01), The low-dose,middle-dose and high-dose groups of BST serum containing can down regulate AQP3 expression level, they are dose-effect in relationship.Conclusion: BST serum containing can influence gene transcription and translation process of AQP3 HT-29 cells,down regulate AQP3 gene mRNA and protein expression level. This suggests that BST may act on the regulatory pathway of AQP3 to regulate the role of water metabolism in vivo,inhibit colonic water absorption, increase the moisture content of the colon content,which may be one of the mechanisms of BST in the treatment of chronic constipation.
