Objective:Identify different metabolites in lymph node metastasis of colorectal cancer through testifying the metabolites and analyzing the clinic materials before making an end-stage model, which can be used to predict the lymph node metastasis. Study the prognostic impact of metabolites on patients in different lymph node metastasis stages based on the metabolites types and levels. It is expected to find a molecular marker to predict the trend of lymph node metastasis for colorectal cancer patients and be reference for judging the tumor stage of colon cancer before operation. Method:1、LC-MS analysis: detect the supernatant of patients in different lymph node metastasis stages by using ultra-performance liquid chromatography mass spectrometry(HPLC-MS) to get the response value of metabolites. Then process the data with accounting software.2、Data processing: process the data with principal component analysis(PCA), t detection, SAM and hierarchical cluster analysis(HCA) respectively to identify different metabolites and further verify them. Results:Results are achieved through the study:1、The PCA shows that different metabolites can distinguish the patients in different lymph node metastasis stages.2、The t detection and HCA show that 17 substances can be find and used to category the patients.3、Discriminate analysis is good for setting a sound prediction model. 10 substances are used in the experimental group and control group, showing good results in preciseness, specificity and sensitivity.4、Logical regression model and end-stage model mapped based on 3 metabolites and clinic pathological parameters are good predictor for lymph node metastasis status.5、Two of the three metabolites have prognostic impact on colorectal cancer patients who are in different lymph node metastasis stages. Furthermore, the differ in response value, the differ in prognostic impact. Conclusion:1、Colorectal cancer patients in different lymph node metastasis stage produce different metabolites.2、Micro-molecular marker can well distinguish the lymph node metastasis status.3、Micro-molecular marker can well distinguish the lymph node metastasis status.4、The selected metabolites can be the signals for the health condition of colorectal cancer patients.
