Aims:we can determine the change of transdermal absorbed dose of bebeerines hydrochloride in different ultrasound power, ultrasound frequency and ultrasound time, with or without chemical penetration enhancers. The studying of the effects of the above factors on transdermal absorption will provide a scientific basis for the setting of parameters (ultrasonic time, ultrasonic power and ultrasonic frequency).Methods:The excised back skin of SD male rats can be used to conduct the experiment of Transdermal drug delivery, by Fixed contact method we can determine The transdermal absorbed dose of Cortex Phellodendri cream in different ultrasound power (low ultrasonic power (0.1w/cm2), medium ultrasonic power(0.4 w/cm2),high ultrasonic power(0.8 w/cm2))in different ultrasound frequency((300KHz,500 KHz,700 KHz,900 KHz,1100KHz) in different ultrasound time (5mins, 10mins,15 mins,20mins)(1) The Transdermal absorbed dose of bebeerines hydrochloride in the condition of chemical penetration enhancers was obviously higher than that in the experimental condition of absence of phonophoresis. The two experiments were in the same experimental condition of absence of phonophoresis. (P=0.019<0.05)(1) The experiment 1 is conducted in the condition of chemical penetration enhancers, while the experiment 2 is conducted in the condition of absence of chemical penetration enhancers. The two experiments are in the same condition of phonophoresis. The accumulated absorbed dose of bebeerines hydrochloride in the experiments lwas obviously higher than that in the experiments 2. (P=0.000<0.01)(1) the difference of the Transdermal absorbed dose of bebeerines hydrochloride in the low-power group (0.1w/cm2), the medium-power group (0.4w/cm2), high-power (0.8w/cm2) Was significant in the same condition of chemical penetration enhancers and ultrasonic frequency. The Transdermal absorbed dose of bebeerines hydrochloride in the low-power group was higher than that in the medium- power group and high-power.(1) The difference of the Transdermal absorbed dose of bebeerines hydrochloride in the different ultrasonic frequency was extremely unobvious in the same condition of chemical penetration enhancers and ultrasonic power (P=0.000< 0.01). The Transdermal absorbed dose of bebeerines hydrochloride in the ultrasonic frequency 1100KHz was higher than that in the other ultrasonic frequency.(1) The effect of interaction between ultrasonic power and ultrasonic frequency on the transdermal accumulated absorbed dose of bebeerines hydrochloride is studied by two - factor variance analysis。It showed that: the effect of Ultrasound power and Ultrasound frequency on the accumulated absorbed dose of bebeerines hydrochloride was significant. (P=0.000< 0.01) it also showed that the effect of interaction between Ultrasound power and ultrasonic frequency on the accumulated absorbed dose of bebeerines hydrochloride was extremely significant(1) The transdermal absorbed dose of bebeerines hydrochloride was continually increasing in 5 minutes,10 minutes,15 minutes,20 minutes, but the difference was not significant. In conclusion:(1) The chemical penetration enhancers can significantly promote transdermal penetration of barbering hydrochloride in Cortex Philodendri.(1) Sonophoresis has a significant role in promoting transdermal penetration of berberine hydrochloride. The absorption.of Berberine hydrochloride is selective for ultrasonic power and ultrasonic frequency. The transdermal accumulated absorbed dose of bebeerines hydrochloride was maximum in ultrasonic frequency of 1100 KHz in the condition of low ultrasonic power (0.1 w/cm2) and medium ultrasonic power (0.4 w/cm2). The transdermal accumulated absorbed dose of bebeerines hydrochloride was maximum in ultrasonic frequency of 500 KHz in the condition of high ultrasonic power (0.8 w/cm2)(1) The Transdermal absorbed dose of bebeerines hydrochloride in Phellodendri cream was the highest in the ultrasonic frequency (0.1w/cm2) in the ultrasonic power 1100 KHz.
