In this experiment, we use Rhamnus bungeana J.Vass.leaves as th e tested material, and use ethanol as the extraction of total flavonoid s. Based on the single-factor tests, the ultrasonic extraction conditio ns of total flavonoids from Rhamnus bungeana J.Vass. leaves were optimized by response surface methodology in order to increase the extraction rate of total flavonoids. Meanwhile, the antioxidant activit y of total flavonoids was assessed by scavenging assays of hydroxy 1 and superoxide anion free radicals. The main results are shown b elow:(1) Firstly, this experiment had extraction of flavonoids from Rhamnus bungeana J.Vass. leaves, grape skin, Sedum spectabile leaves and Ricinus communis L. leaves. Experimental results show that the content of total flavonoid from Rhamnus bungeana J.Vass. leaves is much higher than other materials. Secondly, using different solvents(such as methanol, ethanol, chloroform) to extract, ethanol extract. Therefore, in this experiment, we choose Rhamnus bungeana J.Vass. leaves as experimental materials, and to determine the total flavonoids in ethanol as the extraction solvent.(2)The optimal extraction conditions of total flavonoids were ethanol concentration of 80%, material-liquid ratio of 1:40.Under the optim al conditions, the maximum extraction rate of total flavonoids was 6.11%.(3)The antioxidant activity of total flavonoids was assessed by scav enging assays of hydroxyl and superoxide anion free radicals. As a result, extracted total flavonoids had strong scavenging capacity on hydroxyl and superoxide anion free radicals in a dose-dependent m anner.(4) In the optimum conditions of extraction, macroporous resin purific ation. The results showed that the optimum conditions for adsorption a nd desorption, the concentration of srthe was 5mg/ml, the suitable p H value is 6, the flow rate was 1 ml/min,80% ethanol as elution s olvent,elution rate was 1.2 mL/min, the purity of flavonoids was 34.6 % under the conditions of the above. The content of flavonoids was almost 6 times higher(5)We establish of the evaluation system of antioxidant activity, with rutin and BHT(2,6-butylated hydroxytoluene)compared to the measur ed data of antioxidant and analysis of antioxidant properties and utilit y of Rhamnus bungeana J.Vass. leaves flavonoids. As a result, Rha mnus bungeana J.Vass. leaves flavonoids compare to other flavonoid s which has been studied that showed good antioxidant effect. And Rhamnus bungeana J.Vass. leaves flavonoids’ antioxidant effect is better than BHT. Therefore, it proved that Rhamnus bungeana J.Vas s. leaves flavonoids have a great potential in the field of anti-oxidatio n.
