| Scutellaria barbata D.Don is a kind of traditional Chinese medicine and Labiatae Scutellaria plants, its bitter cold in nature, owned by the lung, liver and kidney. It has the effect of detoxification, diuretic swelling, blood stasis and hemostasis and anti-cancer. Scutellaria barbata has been used for treating sore carbuncle swollen poison, snake bites, hepatitis and hepatomegaly disease. This paper studies the process of extraction and purification, quantitative analysis and renal toxicity inhibition of flavonoids from Scutellaria barbata. This dissertation consists of four chapters.The first part:Study on textraction, separation and purification of flavonoids from Scutellaria barbata. Including:1. The flavonoids from Scutellaria barbata were extracted by the ultrasonic extraction method with extraction quantity based. Ethanol concentration, liquid-solid ratio, ultrasonic timeand extraction times were observed to on the extraction rate of effect the total flavonoids of Scutellaria barbata. Design-Expert software was designed to create the model. The best experiment technique for analysis of response surfacemethod were that ethanol concentration was 70.61%, the liquid-solid ratio was 12:1, ultrasonic time was 52.22 min, and extracted twice. The extraction rate of flavonoid was 13.11%.2. Three kinds of domestic macroporous resin were used to separate and purified total flavonoids from Scutellaria barbata. in the static adsorption and analytical experiment. The results were that sample solution was 5 mg/mL, the sample rate was 2 BV·h-1, the useing elution was 70% ethanol, the elution flow rate was 1BV·h-1, the elution amount was 4 BV mL. The content of the flavonoids was about 75.49% which was 5.74 times the pre-purification.The second part:Study on quantitative analysis of flavonoids from Scutellariae barbatae.HPLC simultaneous determination of five kinds of flavonoids from Scutellariae barbatae was established. Ultimately the content of 5 kinds of effective components flavonoids of Scutellaria barbata were determined, scutellarin was the highest, content is more than 10%, the lowest is quercetin dehydrate, contains nearly 0.6%. This method meets the requirements, which can be used for the quality control of the effective part of the total flavonoids of Scutellaria barbata.The third part:Studies on inhibitory effect of flavonoids from Scutellaria barbata on cisplatin induced renal injury.Through the determination of serum BUN, Scr, ALB spectrophotometric method as well as the content of MDA and SOD in renal tissue homogenate in content, the pathological changes were observed in the mouse kidney tissue HE staining, the inhibitory effect and mechanism of renal toxicity was investigated. The results show that low, medium and high dose of total flavonoids of Scutellaria barbata have significant improvement in renal tissue damage in mice induced by cisplatin, also can reduce the serum BUN and Scr content, the content of serum ALB, MDA content of renal tissue decreased and resulted in increased SOD activity. The experimental results showed that the total flavonoids of Scutellaria barbata can relieve the renal injury induced by cisplatin, and its mechanism may be related toreduced renal oxidative damage is related to the strength.The fourth part:Studies on inhibitory effect of scutellarin and quercetin dihydrate on cisplatin induced renal injury in mice.The mice kidney pathological detection, kidney coefficientand detection of blood urea nitrogenwere (BUN), serum creatinine (SCr), albumin (ALB) changes can be reflected in cisplatin induced renal injury, using biochemical determination of kidney tissue malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) canreflect the oxidative damage. Experimental results show that quercetin dihydrate and scutellarin can improve the renal toxicity. We can also learn that, under the same conditions, quercetinon scutellarin has better effect. |
