| Objective: To explore the profile of urate transporter gene which from ABCG2ã€LGALS9ã€ABCC4ã€PDZK1ã€GCKRã€LRRC16Aã€SLC2A9ã€SLC5A8ã€SLC5A12ã€SLC13A2ã€SLC13A3ã€SLC17A1ã€SLC17A3ã€SLC16A9ã€SLC22A6ã€SLC22A7ã€SLC22A8ã€SLC22A11ã€SLC22A12ã€SLC22A13 in human peripheral blood mononuclear. In further,to explore the possible role of urate transporter gene in PBMCs in gouty patients via the correlation between the expression level of urate transporter gene transcript and the clinic examination indexes.Method:(1) 40 gouty arthritis(20 acute gouty arthritis(AGA), 20inter-critical gouty arthritis(IGA))and 20 healthy controls(HC) were enrolled. Fasting blood samples were obtained for biochemical examination and blood cells counts.And human normal kidney,small intestinal and liver tissue were enrolled as positive control.(2)The expression of urate transporter gene were observed in PBMCs with RT-PCR.(3)The expression of urate transporter gene transcript was detected with RT-q PCR in PBMCs. Correlations between laboratory data and urate transporter gene transcript levels were analyzed by the Spearman’s analysis. In addition,Correlations among all urate transporter gene which were observed in PBMCs transcript level were analyzed by the Spearman’s analysis.Results: 1.Seven urate transporter genes(ABCG2, LGALS9, ABCC4,PDZK1, LRRC16 A, SLC2A9 and SLC17A3) expression were observed inPBMCs both in gout patients and healthy control. 2.Significant increases in the levels of ABCG2ã€LGALS9 mRNA were observed on the PBMCs of gout patients compared with those of healthy subjects(P<0.05, respectively),while significantly decreased in ABCC4ã€SLC2A9 and SLC17A3 mRNA(P<0.05, respectively). No significant difference was observed with respect to PDZK1 and LRRC16 A mRNA between gout patients and healthy subjects(P>0.05, respectively). Significant increases in the levels of ABCG2 ã€LGALS9 mRNA were observed on the PBMCs of acute gouty arthritis and inter-critical gouty arthritis compared with those of healthy subjects(P<0.05, respectively), while no significant difference was observed with it between AGA and IGA(P>0.05, respectively). Significant decreased in the levels of ABCC4 mRNA were detected on the PBMCs of AGA and IGA compared with those of healthy subjects(P<0.05), while no significant difference was observed with it between AGA and IGA(P>0.05). No significant difference was observed with respect to PDZK1 and LRRC16 A mRNA on the PBMCs of AGA,IGA and healthy subjects(P>0.05,respectively),while significant decreased in the levels of LRRC16 A mRNA were detected on the PBMCs of AGA compared with those of IGA(P<0.05).Significant decreased in the levels of SLC2A9 〠SLC17A3 mRNA were detected on the PBMCs of AGA and IGA compared with those of healthy subjects(P<0.05), while significant increased in the levels of SLC2A9 mRNA were detected on the PBMCs of AGA compared with IGA and significant decreased in the levels of SLC17A3 mRNA were detected on the PBMCs of AGA compared with IGA.3. In the GA patients, the levels of ABCG2 mRNA was correlated positively with blood uric acid concentration levels(r=0.24, P<0.05) and the levels of ABCC4 mRNA wascorrelated positively with blood triglycerides levels(r=0.308,P<0.05),while the levels of SLC17A3 mRNA was correlated negatively with lymphocyte levels(r=-0.354, P<0.05).4.In the GA patients, the levels of ABCC4 mRNA was correlated positively with LRRC16Aã€SLC17A3mRNA(P<0.05),while in the healthy control,the levels of ABCC4 mRNA was also correlated positively with LRRC16 A mRNA(P<0.05). Addition,In the GA patients, the levels of LRRC16 A mRNA was correlated positively with PDZK1ã€SLC17A3 mRNA(P<0.05),while in the healthy control,the levels of LRRC16 A mRNA was also correlated positively with PDZK1ã€SLC17A3 mRNA(P<0.05).Conclusion: Only seven urate transporter genes are expression in PBMCs. Altered expression of urate transporter genes in PBMCs might be involved in the pathogenesis of gout. |
