| Objective: Chronic mountain sickness(CMS) is mainly due to high-altitude hypobaric hypoxia. Hypobaric hypoxia can cause multiple organ damage in the body by causing pulmonary arterial hypertension. In this study, the plain group was normal control,the drug groups with angiotensin receptor blocker irbesartan intervention. To investigate the preventative effect of irbesartan on CMS rats with abnormal blood indexes and multiple organ injury. Methods: 120 experimental rats were randomly divided into six groups(plain group, plateau model group, nifedipine group, irbesartan-low-dose group,irbesartan-medial-dose group and irbesartan-high-dose group), then placed in hypobaric oxygen chamber for 30 days and administered drug interventions for 15 days except plain group. After 45 days, pulmonary artery pressure, blood oxygen pressure and saturation of blood oxygen were measured. Furthermore, the effect of irbesartan on the CRP, ET-1, NO,IL-6 in serum and SOD, MDA, GSH-PX in lung tissue homogenates, pathological changes of multi-organ(heart, brain, lung and liver) and heart B ultrasonic were analyzed.Results: It restored the pulmonary artery pressure, blood oxygen pressure and saturation of blood oxygen that bore angiotensin receptor blocker irbesartan, the content of MDA was deceased, while the content of SOD, GSH-PX were increased in lung tissue homogenates. In addition, the content of ET-1, CRP, IL-6 were deceased, while the content of NO was increased in serum. The effect of irbesartan was most significant in the high-dose group. Two-dimensional ultrasound examination showed that irbesartan was effective in the treatment of left ventricular diastolic dysfunction, which was confirmed by pathologic examination. Furthermore, irbesartan in brain, lung and liver tissue injury have improved the efficacy. Conclusion: Angiotensin II receptor blocker irbesartan can improve the effects of CMS rats’ abnormal blood indexes and multiple organ damage through have an impact on pulmonary artery pressure, oxidative stress and inflammatory response,so a new therapeutic direction for the treatment of CMS was proposed. |
