Objective: To study the chemical constituents of anti-breast cancer polyphenols effective parts of pomegranate peel(PEPP), and optimize the formulation of the PEPP granules. Methods: 1)The petroleum extracts, ethyl acetate extracts, n-butanol extracts and water extracts of PEPP were prepared by liquid-liquid extraction. Silica gel column,octadecylsilyl column polyamide and Sephadex gel chromatography were used to separate the chemical components of each polar extracts, different flow divisions were merged according to the results of thin-layer chromatography(TLC). The compounds were purified by recrystallization and preparative high performance liquid chromatography(pHPLC). Structural elucidation of compounds was carried out according to the physical and chemical properties and spectroscopic data. 2)The single factor experiment design was used to optimize the formulation of the PEPP granules, including the ptimization of the type and amount of diluting agent, the amount of citric acid which was used as pH regulator and the antioxidant. The molding rate, angle of repos, solute rate, dilution rate constant, critical relative humidity(CRH), the area under the moisture absorption curve,and the content of polyphenols of different samples were detected as the index. And three scoring systems(the synthetical scored method, normalized value method and the rank sum ratio method) were used to evaluate the granules. Results:1)Sixteen compounds were isolated from the n-butanol extracts of PEPP. Compared with the control article, the compound(16) was confirmed as punicalagin by high performance liquid chromatography(HPLC). According to the physical and chemical properties and 1H-NMR, 13C-NMR spectroscopy date, six compounds were identified, they were narcissin(2),1-O-glaloyl-α-D-glucos(3), tachioside(6), p-hydroxybenzoic acid glucose ester(7),coumaric acid glucose ester(14),and 2-phenylethyl rutinoside(15). Six compounds wereisolated from the pomegranate peel for the first time. 2)The optimized formulation of PEPP granules was as following: the mass ratio of PEPP- mannitol-citric acid was 100∶50∶1. Conclusion: 1)compounds 2, 3, 6, 7, 14 and 15 are isolated from pomegranate peel for the first time, and the results lay the experimental foundation for further elucidation material basis of PEPP. 2)The PEPP granules prepared by the optimized formulation has good appearance, high molding rate, low moisture absorption and good oxidation resistance. 3)The results of three scoring system are close and consistent,which mean that three scoring systems can be used to evaluated the multi index results.
