Plasma Metabonomics Analysis Of Liver Cancer Patients Of Damp Abundance Due To Spleen Asthenia Syndrome

Objective:This study aimed to explore plasma metabonomic changes in liver cancer patients with damp abundance due to spleen asthenia syndrome of Chinese medicine, and to elucidate the metabolic mechanisms in vivo.Methods:1H nuclear magnetic resonance(NMR) based metabonomic analysis was performed on plasma samples from 30 liver cancer patients with damp abundance due to spleen asthenia syndrome, 30 liver cancer patients without the syndrome, and 40 healthy subjects with the syndrome. Spectrogram integral results were analyzed by orthogonal partial least squares discriminant analysis(OPLS-DA).Results:The results showed that compared with healthy subjects, various amino acids mainly including plasma leucine, valine, isoleucine, histidine, phenylalanine, glutamine, glutamic acid, tyrosine, glycine and alanine of liver cancer patients with damp abundance due to spleen asthenia syndrome were significantly decreased(P<0.05). Lactic acid, creatine, beta glucose, alpha glucose, choline levels in plasma were also decreased(P<0.05). However, plasma very low density lipoprotein cholesterol(VLDL-C), beta hydroxy butyric acid, acetone, acetyl acetate, unsaturated fatty acid increased in liver cancer patients with damp abundance due to spleen asthenia syndrome(P<0.05). Compared with liver cancer patients without damp abundance due to spleen asthenia syndrome, patients with the syndrome presented obvious lower plasma contents of leucine, valine, isoleucine, histidine, phenylalanine, glutamine, glutamic acid, tyrosine, glycine and alanine(P<0.05); lower level of plasma creatine(P<0.05); higher contents of lactic acid, acetone, beta glucose, alpha glucose, unsaturated fatty acid, and etc.(P<0.05). Conclusions:Compared with liver cancer diagnosed as damp abundance due to spleen asthenia syndrome patients and non-damp abundance due to spleen asthenia syndrome patients, there were significant differences in metabolomics indicators. Possible metabolic phenotype of liver cancer diagnosed as damp abundance due to spleen asthenia syndrome patients are leucine, isoleucine, valine, glutamine, glutamic acid, histidine, glycine, alanine, tyrosine, phenylalanine, creatine,creatinine,lactic acid, acetone, alpha glucose, beta glucose,unsaturated fatty acidand. It is inferred that the biological basis of same disease with different syndrome is different on traditional Chinese medicine.It also proof that“same disease with different syndrome”has scientificity and rationality on traditional Chinese medicine. Liver cancer patients of the damp abundance due to spleen asthenia syndrome had significant changed in process of glycometabolism,amino acid metabolism and fat metabolism. It is explain that energy metabolism and protein metabolism are abnormal, meanwhile, glyconeogenesis and immune function presents a state of disorder in the liver cancer diagnosed as damp abundance due to spleen asthenia syndrome patients. Moreover, their prognosis may be worse than that of non-damp abundance due to spleen asthenia syndr ome liver cancer patients.
