[Objective]This project based on nourishing blood treatment method through the use Siwutang intervention COPD airway remodeling rat model of disease and airway remodeling‐related cytokines VEGF expression level of TGF‐beta1and MMP‐9/TIMP‐1balancerole, initially revealed Siwutang delay the the COPD rat airway remodeling mechanism of the pathological changes, to provide new ideas and laboratory research base for Siwutang clinical application.[Methods]1. Copy COPD airway remodeling rat model: Take SPF male SD rats and randomly divided into normal control group, COPD airway remodeling model group, Siwutang high, medium and low dose group; addition to the normal control groupin addition, the remaining groups in the first day of the experiment,14days with intra‐tracheal injection of1mg/kg LPS;28days (except for the first14days) in the first two days of the experiment to the group rats were placed in a homemade plexiglasssmoked inside, injected the cigarette smoke,30min/d;2. Drug intervention: Siwutang high, medium and low doses of the drug intragastric administration on rat model of drug intervention in, a period of30days after the end of modeling.3. Indicators of detection:(1) Observed the behavior of the experimental rats histological changes produced lung tissue biopsy were observed;(2) The rat serum by enzyme‐linked immunosorbent assay to detect VEGF, TGF‐β1, MMP‐9/TIMP‐1levels of.[Results]1. Behavioral observation: Compared with the the COPD airway remodeling model group rats, Siwutang high, medium and low doses of the drug intervention in group of symptoms and signs have been greatly improved in the breeding process, a coat of gloss, the overall situation is better.2. Histopathological changes situation: Lung tissue biopsy can be observed high Siwutang, medium and low dose drug intervention group with COPD airway remodeling model group, pulmonary bronchioles and perivascular inflammatory cells decreased, especially in the high dose group. airway wall structure is more complete, reduce airway epithelial mucus secretions.3. Serum cytokine levels:(1) model serum TGF‐beta1, VEGF content was significantly higher than in normal rats, a significant difference (P <0.01), the ratio of MMP‐9/TIMP‐1serious imbalance model serum (<1), compared to the normal group of close to1:1ratio of difference;(2) four‐Wu‐Tang drug intervention group compared with the model group rats have a downward effect on TGF‐β1, VEGF two indexes (P <0.01); the MMP‐9/TIMP‐1the ratio a significant regulatory role.[Conclusion]1. Siwutang of COPD airway remodeling rat lung tissue bronchial and perivascular inflammatory cell infiltration, a certain improvement in the inflammatory status of the airway.2. Siwutang reduce COPD airway remodeling in rat serum TGF‐beta1, VEGF content, adjust MMP‐9/TIMP‐1imbalance, inhibition of the growth of fibroblasts and endothelial cells, the degradation of collagen synthesis and bronchial wallmatrix deposition and vascular wall, slowing the COPD airway remodeling pathological changes.3. Traditional Chinese medicine nourishing blood method by promoting blood airway remodeling key factor TGF‐beta1, VEGF expression is reduced, adjust the MMP‐9/TIMP‐1balance, thereby preventing pathological proliferation of the extracellular matrix, intervention in COPD airway structureplastic.
