The study on the Principle of reducing toxicity of Processing with Vinegar of Euphorbia ebracteolata Hayata was the State Pharmaceutical Administration special fund public sector projects-made and toxic medicine bran technology and traditional Chinese medicine principles study (NO.200807039) funded projects,was an traditional chinese pharmacology open issue,in advantaged subject construction project in Jiangsu Province (2011ZYX2-011)The process standardization and quality standardization of toxic chinese medicine is the key to ensure the its clinical application safe and effective, and processing mechanism is foundation to judg processing technology and quality standard. Therefore, the processed mechanism of toxic traditional Chinese medicine has very important significance to ensure its safe and effective clinical application.Langdu,which is one of the 2010 edition of "Chinese Pharmacopoeia" re-included varieties, is one of the 28 toxic drugs of national medical use management species announced by Ministry of Health. The main source is Euphorbia ebiacteolata Hayata and Euphorbia fischeriana Steud. It has good effect,but the toxicity of the mouth, throat irritation and congestion burning, vomiting, abdominal pain, diarrhea, bleeding diarrhea stools, and even convulsions, mania and so on. Therefore, it was often used after processed,and the representativeness processing methods was"processed with vinger."Previous study we found that its toxicity mainly as a strong irritant, reproductive toxicity, after processed, the toxicity was significantly weakened. During the study, we also found the following problems:First, the lack of Euphorbia ebracteolata product reference and irritating ingredients is not clear, for follow-up studies difficult. Second, Euphorbia ebracteolata processed mechanism is not clear,and the inadequate research of quality control, so that quality standards can not be perfect, unable to guide clinical medication safety. Consequently, Euphorbia ebracteolata irritating ingredients and processing mechanism of attenuation is particularly urgent. In response to these problems, the subject has an research on Euphorbia ebracteolata irritating parts、the change in HPLC fingerprints and chemical composition before and after processing. And we also find A novel strategy to rapidly explore potential chemical markers for thediscrimination between raw and processed samples by UHPLC-TOFMS with multivariate statistical analysis.The main concents as follows:1 Literture researchSummarized the textual and processing history of langdu,and the chemical composition、 pharmacological efffects and clinical applications of Euphorbia ebracteolata Hayata, laid a theoretical foundation for the further study of Euphorbia ebracteolata Hayata.2 Irritating part researchThe extraction method and the Irritating part was investigated as using the gastrointestinal irritation experimental of mouse.And there are six compounds was separated from the irritating part:jolkinolide B, β-amyrinacetate,2,4-dihydroxy-6-methoxy-3-Methy-1-acetophenon-e,24-methylenecycoartanol and ebractedatanolide A. Provide a reference substance for Euphorbia ebracteolata further study.3 HPLC fingerprints research10 batches of Euphorbia Ebracteolata Hayata hygiene products and 10 batches of processed products were selected from different origin. Acetonitrile and water as mobile phase (gradient elution) was used to establish HPLC fingerprints of crude and processed Euphorbia Ebracteolata Hayata. the characteristics of Euphorbia Ebracteolata Hayata hygiene products have been compared with processed products in the study.It provides effective methods for the quality control of Euphorbia Ebracteolata Hayata. The main mechanisms underlying herb processing may be related to the changes in the content of the components in the herbs, and we should have a continued study.4 The chemical change researchTo investigate the change of 3 components with different methods and temperature,and develop the HPLC specific chromatogram,the content of ebracteolata cpd B、ebracteolata cpd C and jolkinolide B in Euphorbia Ebracteolata Hayata processed with different methods and temperature were analyzed by HPLC method, and the HPLC specific chromatograms were established. The content of active constituents ebracteolata cpd B、ebracteolata cpd C increased firstly and then decreased as the heating temperature increased. Different process materials and temperature had different influence on the Euphorbia ebracteolata Hayata.Sauteing with vinegar can give full play to the role of reduce toxicity compared to the other methods. The processed vinegar could not be replaced by acetic acid.5 The characteristical marker researchAn approach using UHPLC-TOFMS along with multivariate statistical analysis was developed to rapidly find potential chemical markers.We use UHPLC-TOFMS to determine batchs of raw and processed products,and then use multivariate statistical analysis to subject the datasets of tR-m/z pair, ion intensity and sample code to holistically compare the difference between raw and processed samples. The components that correlate to these ions should be the processing-induced transformed components, and could be regarded as potential chemical markers for evaluating the extent and investigating the mechanism of processing.
