ObjectivePrevious researches suggested that most alkaloids which were isolated from Erythrina plants showed simplex structure type of isoquinoline alkaloids. However, our group have found some new skelecton Erythrina alkaloids from Erythrina variegate, which cause our attention on this genus plants. We carried out the alkaloid chemical investigations to explore more compounds with novel structure and biological activity in this genus. Futhermore, bioactivity investigations provide the basis for the comprehensive development and utilization of E.arborescens Roxb.MethodA systematic method of phytochemical research is used to isolate alkaloids constituents of E. arborescens Roxb. stems and leaves, which includes the extraction, alkali treatment, normal and reversed phase chromatographic separation. Structures were determinated by NMR, UV, IR, and mass spectrometry methods. The activity of new compounds were screened by the methods of MTT, DTNB, and Luciferase assay, respectively, evaluate the activity of cytotoxic and anti-acetyl-cholinesterase, the influence of Nf-KB signal pathway.ResultsThirty three alkaloids, including fifteen new alkaloids were isolated from E. arborescens Roxb. These new alkaloids were identified as Erythriarborine A (1), Erythriarborine B (2), Erythriarborine C (3), Erythriarborine D (4), Erythriarborine E (5), Erythriarborine F (6), Erythriarborine G (7), Erythriarborine H (8), Erythriarborine I (9), Erythriarborine G (10), Erythriarborine K (11), Erythriarborine L (12), Erythriarborine M (13), Erythriarborine N (14), Erythriarborine B (15). In addition, Erythriarborine H (8) was a dimer Erythrina alkaloid. However, the bioactive screening didn’t disclose active compound.ConclusionsE. arborescens Roxb. is rich in Erythrina alkaloids with structural complex. However, their bioactivity need more wide and futher research.
