Experimental Study Of Herba Epimedii And Fructus Ligustri Lucid On The Rat Asthma Model With Glucocorticoid Aerosol Inhalation Based On The Expression Of GR And HSP90

Objective Based on the expression of GR and HSP90 to study Herba Epimedii and Fructus Ligustri Lucid on the rat asthma model with glucocorticoid aerosol inhalation.Methods105 male SD rats were randomized into seven groups: normal group,model group,glucocorcoid group,YN group,YN combinate glucocorcoid group, YN active ingredients group,YN active ingredients combinate glucocorcoid group. Except normal group,all of the group are Intraperitoneal injection and Atomization inhalation with OVA for five weeks to build asthmatic rat model and glucocorcoid group treated with 0.01% budesonide 30 min,YN gorup treated with Herba EpimediiFructus igustrilucidi,YN combinate glucococoid group treated with both of Herba EpimediiFructus igustrilucidi and 0.01% budesonide 30 min.YN active ingredients group treated with Herba EpimediiFructus igustrilucidi active ingredients, YN active ingredients combinate glucocorcoid group treated with YN active ingredients and 0.01% budesonide.While normal group and model group treated with normal saline to set up contrast.Get blood and bronchoalveolar lavage fluid on day 65. Using HE、masson and PAS staining method to detect lung tissue pathology change.Using Flow cytometry to detect The lymphocyte apoptosis rate and contnet of GR and the binding force of GCR. Using Immunohistochemical to detect contnet of HSP90 in lung tissue and BALF. Using Immunohistochemical,Immunofluorescence and western blot to detect the contnet of GRα,GRβ in lung tissue.Using Fluorescence quantitative PCR to detect the contnet of HSP90、GR、GRα、GRβ mRNA in lung tissue. Calculating the ratios of protein and Mrna of GR and HSP90.ResultsCompared with normal group,The value of Wam,Wat, goblet cell, collagen fiber in lung tissue and the serum level of IL-4,IgE in asthma model group were increased significantly(P<0.01 or P<0.05). The level of lymphocyte apoptosis rate in BALF was decreased significantly(P<0.05). the contnet of GR,the binding force of GCR in BALF and the expression quantity of protein and mRNA of HSP 90, GRα and GRβ were decreased significantly(P<0.01 or P<0.05). GRα/HSP90 protein and mRNA,GR/HSP90 mRNA show imbalanced(P<0.01 or P<0.05). After administration of Herba Epimedii and Fructus Ligustri Lucidi decoction, the content of Wam,Wat, goblet cell, collagen fiber in lung tissue and the serum level of IL-4,IgE showed a clear upward trend(P<0.01 or P<0.05).The level of lymphocyte apoptosis rate in blood and BALF(P<0.01 or P<0.05), the contnet of GR,the binding force of GCR in BALF and the expression quantity of protein and mRNA of HSP 90, GRα and GRβ showed a clear downward trend(P<0.01 or P<0.05). Improve the imbalance of GRα/HSP90 protein,GR/HSP90、GRα/HSP90、GRα/ GRβ mRNA(P<0.01 or P<0.05).Conclusions1.The mechanism of asthma maybe the airway inflammation, airway remolding, reduction of GC sensitiveness and GR/HSP90 imbalance.2. Herba Epimedii and Fructus Ligustri Lucidi play a role in asthma treating through the following ways:Reversing airway remodeling,inhibiting the inflammation reaction of lung tissues and enhancing the susceptibility of hormones effect.3.The good effect in prevention and treatment of asthma of the compatibility active ingredients is equal as the compatibility decoction. Specifically, the compatibility active ingredients groups not only has no difference with the compatibility decoction group, but also has a more significant improvement in some of the indicators.4. The mechanism of the compatibility decoction and its active ingredients with glucocorticoid for treating asthma is related to the following aspects: alleviating the inflammatory reaction, promoting lymphocyte apoptosis rate,increasing the the expression quantity of protein and mRNA of HSP 90 and GR to enhance the susceptibility of hormones effect. Improve the imbalance of GRα/HSP90 protein,GR/HSP90、GRα/HSP90、GRα/ GRβ mRNA.
