Abalone is famous for its delicacy and curative effect on some disease.The by-products from the processing of abalone,viscera may pose a disposal problem due to the ease of spoilage and environmental pollution.To increase the market value of the discards,the polysaccharide was extracted and purified from the viscera of abalone,Haliotis Discus Hannai Ino.The bioactivity of the polysaccharide was also investigated.The results of antioxidant activity in vitro showed that,pepsin was the best protease to deal with the crude abalone viscera polysaccharide(CAVP),the optimal usage of pepsin for female and male viscera was 2000 U/g and 4000 U/g,respectively.After treatment with pepsin,the hydrolysate of CAVP showed notable scavenging activity on hydroxyl radical,higher than that of Vc,but relatively weak reducing power and chelating ability.There is no correlation between the polysaccharide or protein contents with the antioxidant activity.Two purified polysaccharide fractions were separated by Sephadex G-100 column after the CAVP being treated with pepsin,named as AVPⅠand AVPⅡ.The antioxidant of AVP, AVPⅠand AVPⅡextracted from female and male abalone showed also most the same rule. The capability of scavenging hydroxyl radical was AVP>AVPⅡ>AVPⅠ,reducing power was AVPⅡ>AVP>AVPⅠ.But the chelating ability of AVPⅠand AVPⅡcould not be detected.The molecular masses of AVPⅠand AVPⅡwere 272 kD and 6 kD.The polysaccharide contents of AVPⅠand AVPⅡwere 66.41%and 51.84%,and the protein contents were 1.70% and 15.55%.AVPⅠand AVPⅡshared the same monosaccharide as rhamnose,glucose,xylose, mannose,galactose and fucose.The ratio for AVPⅠwas 1:2.15:4.00:5.36:33.18:45.14, whereas AVPⅡwas 1:1.46:1.33:4.98:16.08:12.51.AVPⅠand AVPⅡcould restrain the proliferation of Hela and K562 cell in vitro. Moreover,it was found that AVPⅠand AVPⅡcould dose-dependently induce a significant increase in spleen and thymus weight,lymphocyte proliferation,phagocytosis of macrophage, natural killer cell activity,antibody production in normal and CTX-caused immunosuppression mice.AVPⅠand AVPⅡcould not only improve the immune system of normal animal.Moreover,they could be used as a antagonize agent against CTX,which was widely used as an antitumor medicine.Thus the two polysaccharides could be developed as an adjuective therapy in tumor healing.The polysaccharide from the viscera of abalone could also be used for industrial purposes.
