Aralia chinensis L.,which belongs to Araliaceae,Aralia,was edible and medicinal plants,This plant had abundant wildlife resources,with analgesic anti-inflammatory,blood stasis,diuretic,stomach and efficacy of hypoglycemic.Root bark and stem bark were used medicinally.The root bark of Aralia chinensis polysaccharide?ACP?was one of the effective natural constituents of Aralia chinensis.Extraction process,molecular weight determination and monosaccharide composition of ACP were analyzed by study.It provided a theoretical basis for better exploitation of ACP.The main experiment results were as follows:1.The extraction methods and conditions of ACP: The polysaccharide yield of the root bark of Aralia chinensis was used to measure the extraction efficacy.The optimal single factor experiments,by orthogonal design optimization of hot water extraction ACP as follows: extraction time was 210 min,extraction temperature was 80 ?,solid-liquid ratio was 1:30?g/mL?,under these conditions the polysaccharides extraction rate was 8.334%.The optimal single factor experiments,by orthogonal design optimization of ultrasonic-assisted extraction ACP as follows: extraction time was 80 min,extraction temperature was 70 ?,solid-liquid ratio was 1:30?g/m L?,ultrasound power 100 W,under these conditions the polysaccharides extraction rate was 11.133%.The optimal single factor experiments,by orthogonal design optimization of microwave-assisted extraction ACP as follows: extraction time was 40 min,extraction temperature was 50 ?,solid-liquid ratio was 1:30?g/mL?,microwave power 400 W,under these conditions the polysaccharides extraction rate was 15.189%.Compared with conventional water extraction,ultrasound-assisted extraction and microwave-assisted extraction greatly shorten the extraction time,increasing the extraction yield of polysaccharides.2.After the removal of protein and dialysis treatment,ACP protein removal rate was 70.060% and polysaccharides reserved were 59.172%.by using coomassie brilliant blue method.The ultraviolet spectrum analysis showed that no obvious absorption peak of 270 290 nm.Indicating the sevage method is good for root bark of ACP protein impurity removal effect.3.ACP molecular weight was measured,using SEC-LLS measurement method,weight-average molecular weight?Mw?of ACP was 1.414×106g/mol,number average molecular weight?Mn?was 5.259 × 105 g/mol,molecular weight distribution index?Mw/Mn?was 2.688,indicating that the molecular weight distribution is narrow;chromatography figure shows a single symmetrical peak,indicating that the uniformity of the polysaccharide sample.Characterized by infrared spectroscopy shows that ACP had polysaccharide absorption peak(3419.78 cm-1,2926.01 cm-1,1651.06 cm-1,1419.60 cm-1,1024.19 cm-1).4.Using GC-MS analysis monosaccharide composition of ACP after hydrolysis and derivatization,according mixed monosaccharide standard chromatogram,the ACP was composed of D-arabinose,D-glucose,D-galactose,the relative molar ratio: 4.67: 76.44: 18.18.
