| Antler is rich in protein,and it contains 7 kinds of essential amino acids.It has the effects of warming the kidney-yang,strengthening bones and muscles,promoting blood circulation and detumescence.Antler so it has the applied value for developing its peptides.In this article,samples are taken as the antlers of Cervus elaphus provided by Guangfeng deer products trade limited company of Tieling Liaoning.Studies are made into the extraction of protein from the antler of Cervus elaphus,enzymatic hydrolysis processes as well the anti-oxidization,anti-fatigue performance,the regulating immune response and acute toxic safety inspection.The following accomplishments are made:Determination of polysaccharides and protien in antlers of Cervus elaphus:The content of total protein from antler and protein extractive were determinanted.Adjust the protein extractive concentration as 100 mg/mL,and the polysaccharide content in it was tested.The results showed that the total content of protein was 37.5%,and the polysaccharide content was 0.14%.The best hydrolysis process about the Anlter Protein of Cervus elaphus:Through Single-factor test,the Protamex and flavourzyme were selected from proteinases.The protain extractive with the congcentration of 100 mg/mL was treated by Protamex and flavourzyme.The results showed that the optimum condition of protamex for hydrolysis antler protein was pH 7.0,4 h,enzyme concentration 6500 U/g,55 ℃,and the flavourzyme was pH 7.0,3 h,enzyme concentration 750 U/g,45 ℃.The hydrolysis degrees are 33.50%and 19.61%.With the optimum condition conditions,the combination zymohydrolysis for anlter protein,added protamex first(pH 7.0,time 4 h,temperature 55 ℃,enzyme quantity 6500 U/g,the enzyme is active)and then added flavourzyme(pH 7.0,time 3.5 h,temperature 45℃,enzyme quantity 850 U/g),had best effect on digesting antlers protein,and the hydrolysis degree was 53.77%.The antioxidant actibity of the polypeptide and the protein was studied by the deoxidizational ability,the scavenging capacity of O2-·and ·OH.The antioxidant actibity in vivo was studied by SOD and MDA levels in mice.All the results show that the Cervus elaphus’s Anlter protein has great antioxidant ability,and polypeptide’s anti-fatigue ability is more apparent.The antioxidant activities in vitro of the Cervus elaphus’s antler peptides and protein with the same concentration had extremely distinct difference(P<0.01).In the tests of antioxidant activities in vivo,SOD activity in mice of the groups with Cervus elaphus’s antler peptides compared to the protein,the difference was significant(P<0.05).While the MDA content,the difference between groups was extremely significant(P<0.01).The anti-fatigue ability of the polypeptide and the protein is looked into with the mice’s loaded swimming time and its level of liver glycogen and blood lactic acid as criteria.Tests results show that Cervus elaphus’s antler polypeptide and protein has anti-fatigue ability.Thus,the mouse’s loaded swimming time can increase significantly.This shows that the polypeptide of Cervus elaphus’s antler polypeptide has great anti-fatigue ability.Compared to the group of Cervus elaphus’s antler protein,loaded swimming time of mice in the group of peptides and the contents of Liver Glycogen were extremely significantly increased(P<0.01).The content of blood lactate of peptides group were significant different(P<0.05).The immune activity of the antler polypeptide and the protein was studied by the capacity of lymphocyte proliferation induced and the indexes of the clearance rate of carbon particles.The results show that the polypeptide and the protein of Cervus elaphus’s antler has great ability to improve the immune function of mice.The index a of Carbon clearance and splenic lymphocyte proliferation in groups of peptides had distinct difference(P<0.05),compared to the Cervus elaphus’s antler protein.The result of acute toxicity testing shows LD50>10 g/kg,so we can say that these Cervus elaphus’s Antler polypeptide are all innocuity,and they are safe to eat.The organs such as:heart,liver,lung,kidney,stomach in mice which are treated by Cervus elaphus’s anlter polypeptide had no significant differences with the control group。which implied that the Cervus elaphus’s anlter polypeptide had no bad influence on the growth of the mice.This study lays the foundation for the high value use of the Cervus elaphus’s antlers and also provides a theoretical basis for the deep processing of Cervus elaphus’s antler. |
