Abalone as one of the sea treasures is popular by everyone.Today,mainly to the cultivation,making abalone food and use more common.However,due to the extremely easy to corrupt seafood,abalone processing and transportation inconvenience.In this paper,the storage process of abalone as the research object,respectively for cold storage and room temperature storage of the total bacterial DNA extraction of abalone.The abalone during storage of bacterial diversity and structure by using PCR-RFLP method and high-throughput sequencing.The conclusions were as follows:1.The diversity of abalone during normal temperature and cold storage was analyzed by PCR-RFLP.A total of five bacterial species were identified,namely,Proteobacterium,Bacteroides,Fusobacteria,Firmicute and Actinomycete;19 bacterial species,respectively,Psychromona,Vibrio,Lactococcus,Lactococcus,Flavobacterium and Fusobacterium.At the cold storage(4 ?),Proteobacteria were used to isolate the dominant bacteria and Bacteroides did not increased in storage time.The species of Vibrio and Arcobacte were first increased and then decreased.The changes of bacteria were not obvious at 12 days and 18 days after storage.The dominant bacteria at 4 ? for 18 days were 5 strain Fusobacteriaceae(29%).The dominant bacteria for 72 h at 20 ?were 4 strain Lactococcus(30%).2.On the basis of PCR-RFLP method,the bacterial species of abalone stored under the cold storage and at room temperature,this experiment used abalone which storage at 4 ? determined by high throughput sequencing.Analysis of bacterial diversity in abalone(4 ?)by high throughput method.A total of 12 bacterial phyla,were Proteobacteria,Bacteroidetes,Fusobacterium,Tenericutes,Deinococcus-Thermus,Acidobacteria,Verrucomicrobia,Spirochaetes,Cyanophyta,Fibrobacteres,Actinobacteria and Firmicutes.The species rich bacterial species for the top ten:Vibrio Psychrilyobacter,Shewanella,Mycoplasma,Crocinitomix,Photobacterium and Polaribacter,Methyloversatilis,Pseudomonas,Pseudoalteromonas.The dominant bacteria were Od(45.2%,Bacteroidetes,and the dominant bacteria were Vibrio,2.9%at the 12d.3.Based on the analysis of the diversity of bacteria during the storage of abalone,the bacterial structure of abalone was added.It provided the theoretical basis for the process of abalone processing,storage,transportation and so on,which provided the theoretical basis for the spoilage and flavor changes caused by bacteria.
