Selaginella Pulvinata belongs to Selaginella genara of Selaginellaceae family, which has activity of anti-diabetics, anti-viral, anti-tumor, hemostasis and immune regulation. It’s been used in folk as a medicine against diabetes and cancer. Potentilla Discolor Bunge (PDB), one of the plant of Potentill genus, has antidiarrheal, hypoglycemic and anti-tumor effects. However, there are separation difficulties in process of separating bioactive compounds from S. Pulvinata and PDB. The thesis aims to develop a fast and efficient method to separate pure compounds from the two plants.The S. Pulvinata was extracted by 95% ethanol. Four biflavones and seven acetylenic phenols were isolated by numerous separation methods, such as organic solvent extraction, silica gel column chromatography, macroporous resin, preparative high performance liquid chromatography and LC-MS. The structure of these compounds were identified by NMR, MS and some other analytical tools, and they were hinokiflavone(SPO1), isocryptomerin(SP02), sotetsuflavone(SP03), amentoflavone(SP04), selaginellin(SP05), selaginpulvilin A(SP06), selaginpulvilin B(SP07), selaginellin O(SP08), selaginellin E(SP09), selaginellin A(SP10), selaginellin B(SP11).The dry powder of PDB was processed by polyamide column chromatography, silica gel column chromatography, recrystallization, pHPLC and other methods. Finally six compounds were separated and their structure were identified as kaempferol-3-O-(6"-O-E-p-coumaroyl)-β-D-glucopyranoside(PD01), apigenin(PD02), maslinic acid(PD03),3’-O-methylellagic acid-4-O-β-D-glucopyranoside(PD04), β-daucosterol(PD05), apigenin-7-O-β-D-glucuronoside (PD06). Compound PD04 was isolated from PDB for the first time.The compounds SP01 to SP11 were evaluated for their in vitro inhibition effect against tumor cell NSCLC H322 via MTT assay. The results showed that all compounds except compounds SP04 and SP07 have activity against NSCLC H322.
