Objective(1)Observing the clinical effect of Potentilla discolor Bunge(PDB)patients with type 2 diabetes,and the influence on tumor necrosis factorα(TNF-α)(2)Observing the effect of PDB flavones on the expression of GULT2m RNA.Exploreing the mechanism of Potentilla discolor Bunge in the treatment of diabetes.Methods(1)Clinical study:60 cases were randomly divided into the treatment group and the control group,with 30 cases in each group.The control group was treated with western medicine,and the treatment group was treated with PDB granules on the basis of the control group.Both groups were observed for8 weeks.Observing the changes of fasting blood glucose(FPG),2h postprandial blood glucose(2h PG),Hb A1c,TNF-αin 2 groups before and after treatment.(2)Experimental study:40 SD rats were randomly divided into blank control group(n=8)and model group(n=32).After modeling,the rats were randomly divided into model group,PDB group and Pioglitazone group.After modeling,the gavage is beginning,the control group,model group,the PDB group(160mg·kg-1·d-1),the pioglitazone group(1.95mg·kg-1·d-1)was correspondingly gave oral medication respectively.Respectively observing the general signs,weight and blood glucose of rats after modeling and with the treatments for 2 weeks and 4 weeks.Testing the expression of GLUT2 in rat livers with fluorescence quantitative polymerase chain reaction(RT-PCR).Results(1)Clinical study:PDB granules combined with western medicine has a good control effect on fasting and postprandial blood glucose,and it has high safety.The level of TNF-αin treatment group was lower than that in control group(P<0.05).PDB have certain effect on blood glucose,which may through regulate TNF-αto reduce the metabolic inflammatory response of the body.And the specific mechanism remains to be fully elucidated.(2)Experimental study:(1)Compared with the model group,PDB can improve the symptoms of increased diet,increased excretion and weight loss in type2 diabetic rats,and improve the condition of less activity in type 2 diabetic rats.(2)PDB could reduce the blood glucose of type 2 diabetic rats,and the difference was statistically significant(P<0.05).(3)RT-PCR results showed that the expression of GLUT2 in the PDB group was significantly higher than that in the model group(P<0.05),which indicated that the PDB may play a role in reducing blood glucose by promoting the expression of GLUT2.Conclusion(1)Clinica trials showed that PDB have a clear effect in the treatment of type 2 diabetes,and it is safe and reliable in improving the efficacy of western medicine.The hypoglycemic effect of PDB may be related to the inhibition of TNF-α.(2)PDB may play a role in decreasing the hypoglycemic by increasing the expression of GLUT2.
