Objective:This study adopts Herbs-partition Moxibusition on Shenque acupoint to treat Primary dysmenorrheal.With dysmenorrheal rat-model established,each group’s metabolites spectrum of urine are observed and tested after treatment.Through identifying potential biomarkers,related metabolic pathways are deduced and then effectivemechanisms of this therapy are explored.Methods:In this study,40 healthy SD rats that were unpregnant female randomly divided into blank group,model group,gastric-lavage group and herbs-partitioned moxibustion group.With treated rationally,each group was compared and observed of incubation period of writhing response as well as writhing times in 30 min.Each group’s urine samples were collected after rat-model established favorably.And their urine metabolites were tested by metabonomics technology named GC-TOF-MS to acquire related metabolites about treatment of Primary dysmenorrheal by Herbs-partition Moxibusition on Shenque acupoint and to deduce related metabolic pathways.With comparing to gastric-lavage group differences about metabolites as well as pathways,bridges between clinical symptoms,characteristics of metabolites,pathways and Shen que acupoint’s distinctive effectivity are build up.Results:1.Metabolites spectrum produced by motherwort compoundtreating Primary dysmenorrheal has changed dramatically.They are mainly appeared as follows:hippuric acid,maltose,sorbose,quinic acid,oxalic acid,gluconic lactone,creatine deaminase,2,6-diaminopimelic acid,dihydroxyacetone and N-epsilon-Acetyl-L-lysine.Among these,hippuric acid involved in phenylalanine metabolism,maltose as well as gluconic lactone referred to glycometabolism,creatine deaminas and2,6-diaminopimelic acid concerning about amino acid metabolism,while other metabolic pathways are not reported yet.2.Metabolites spectrum generated by Herbs-partition Moxibusition on Shenque acupoint treating Primary dysmenorrheal has varied evidently.They are mainly showed as follows:glycine,xylose,citric acid,oxalic acid,sorbose,hippuric acid,benzoic acid,3-(3hydroxypheny)propionic acid,gluconic acid,m-cresol,pantothenic acid,4-oxo-1H-quinoline-2-carboxylic acid,myo-inositol,glycocyamine and gluconic lactone.Among these,hippuric acid,benzoic acid and3-(3hydroxypheny)propionic acid are related to phenylalanine metabolism.Gluconic acid,myo-inositol as well as gluconic lactone are referred to glycometabolism.glycocyamine and4-oxo-1H-quinoline-2-carboxylic acid are involved in amino acid metabolism,meanwhile citric acid is referred to steroid hormone metabolism.However,other metabolic pathways are not informed yet.obviously,the treatment of Herbs-partition Moxibusition on Shenque acupoint treating Primary dysmenorrheal outnumber more metabolism pathways than motherwort compound’s.Conclusions:Metabonomics mechanisms of Herbs-partition Moxibusition on Shenque acupoint treating Primary dysmenorrheal are mainly based on reproductive endocrine hormone(steroid hormonemetabolism),local algogenic factors(glycometabolism),substances associated with immune system(amino acid metabolism)and the regulation of neurotransmitter(phenylalanine metabolism),which influence the body system of nerve,internal secretion,immune system to relieve the pain as a treatment.
