Study On The Prescription Optimization And Pharmaceutics Of Long Chi Tablet

Objective:Long Chi Tablet(LC tablet),consisting of Gymnema sylvestre(Retz.)schult(abbreviated as GS),Dranaene cochinensis(Lour)S.C.Chen(abbreviated as DC)and Glycyrrhiza uralensis Fisch(abbreviated as GU),is a clinical prescription in the People’s Hospital of the Guangxi Zhuang Autonomous Region.This paper studies the acute toxicity,optimal prescription and main pharmacodynamics of LC tablet.Methods:(1)219 times the human dose,i.g,to observe the poisoning symptoms and death in mice.(2)By central composite design-response surface methodology(CCD-RSM)optimization test,we titrated the dosage of GS,DC and GU.(3)The diabetic hyperglycemia models of mice respectively induced by Streptozotoci n(STZ),Adrenaline(AD)and Glucose were employed to study the hypoglycemic effect of LC tablet.(4)Acute blood-stasis model rats induced by swimming in ice water and then being injected AD was employed to observe the effects on hemorheology.Mesenteric microcirculation disorder model rats induced by being injected AD were used to observ the effects on microcirculation.And the blood clotting time were determined to evaluate the effects on PT and APTT in rabbits.Results:(1)219 times the human dose,i.g,poisoning symptoms and death in mice did not occur within 7d.(2)The dosages of GS,DC and GU are positively correlate to the hypoglycemic effect;on the other hand,the dosages of DC is positively correlate to the effect of promoting blood circulation and removing blood stasis.CCD-RSM optimization test gave the rang of GS,DC and GU as:8.17～16.83g,1.21～1.79g,7.42～9.00g,respectively.Based on the clinical experience,we got the optimized prescription as:12.5g for GS,1.5g for DC,and 9g for GU.(3)LC tablet could significantly lower the blood glucose level and improve the hypoglycemia symptoms in hyperglycemia mice respectively induced by STZ,AD and Glucose.(4)LC tablet can significantly lower the animals’ whole blood viscosity,plasma viscosity,blood fibrinogen concentration,ESR and platelet adhesion rate;can improve the hypercoagulable and high viscous state of blood so that avoid the appear of thrombosis;can make improvement in arterial narrowing,slow blood flow,reduction of open capillary and change of blood flow pattern;can extend the PT and APTT time.Conclusion:(1)LC tablet is a novel TCM formula without toxicity.It has the pharmacological effects of lowering blood glucose level and promoting blood circulation and removing blood stasis.(2)The optimized prescription is 12.5g,1.5g and 9g for GS,DC and GU,respectively,which is very similar to the original one.(3)This paper provides scientific underpinning for the new drug development and clinical application of LC tablet.
