The immunohistochemistry and in situ hybridization were used to detect the expression MDR1, MRP1, ABCG2 protein and m RNA in incised margin normal mucosa of colorectal adenocarcinoma, colorectal adenoma and colorectal adenocarcinoma. To investigate the expression difference of MDR1, MRP1, ABCG2 in incised margin normal mucosa of colorectal adenocarcinoma, colorectal adenoma and colorectal adenocarcinoma and relationship with clinical pathological features; the correlation among MDR1, MRP1 and ABCG2 protein and m RNA in colorectal adenocarcinoma; To further realize the mechanism of colorectal adenocarcinoma occurrence, development, and provide valuable reference indicators and experimental basis for clinical colorectal adenocarcinoma chemotherapy and prognostic.Formalin fixed paraffin embedded tissues were collected from January, 2013 to June, 2014 in the First Affiliated Hospital of Hebei North University. There were 116 cases of colorectal adenocarcinoma, 50 cases of colorectal adenoma and 30 cases incised margin normal mucosa of colorectal adenocarcinoma, all colorectal adenocarcinoma without preoperative radiotherapy and chemotherapy. Respectively by mean of immunohistochemistry and in situ hybridization to detection the expression of MDR1, MRP1 and ABCG2 protein and m RNA. Analysis of MDR1, MRP1, ABCG2 protein and m RNA relationship with clinicopathological factors(age, gender, tumor differentiation degree, tumor locations, lymph node metastasis and TNM stage in colorectal adenocarcinoma).The immunohistochemistry results showed that: The positive rates of MDR1 protein were 30%(9/30),10%(5/50),72.4%(84/116) in incised margin normal mucosa, colorectal adenoma and colorectal adenocarcinoma. The expression of MDR1 among groups had statistical difference(P<0.05). The expression of MDR1 protein were related to tumor differentiation degree, lymph node metastasis and TNM stage(P<0.05), but not related to the age, gender and tumor locations(P>0.05). The positive rates of MRP1 protein were 16.7%(5/30),40%(20/50),70.6%(82/116)in incised margin normal mucosa, colorectal adenoma and colorectal adenocarcinoma. The expression of MRP1 among groups had statistical difference(P<0.05). The expression of MRP1 protein were related to tumor differentiation degree, lymph node metastasis and TNM stage(P<0.05),but not related to the age, gender and tumor locations(P>0.05). The positive rates of ABCG2 protein were 43.3%(13/30),12%(6/50),75.6%(88/116)in incised margin normal mucosa, colorectal adenoma and colorectal adenocarcinoma. the expression of ABCG2 among groups had statistical difference(P<0.05). The expression of ABCG2 protein were related to tumor differentiation degree, lymph node metastasis and TNM stage(P<0.05), but not related to the age, gender and tumor locations(P>0.05). Spearman showed positive relative among expression of MDR1, MRP1, ABCG2 protein in colorectal adenocarcinoma(P<0.05). The in situ hybridization results showed that: The positive rates of MDR1 m RNA were 33.3%(10/30),6%(3/50),72.4%(80/116) in incised margin normal mucosa, colorectal adenoma and colorectal adenocarcinoma. The expression of MDR1 among groups had statistical difference(P<0.05). The expression of MDR1 m RNA were related to tumor differentiation degree, lymph node metastasis and Dukes stage(P<0.05), but not related to the age, gender and tumor locations(P>0.05). The positive rates of MRP1 m RNA were 20%(6/30),44%(22/50),74.1%(86/116)in incised margin normal mucosa, colorectal adenoma and colorectal adenocarcinoma. The expression of MRP1 among groups had statistical difference(P<0.05). The expression of MRP1 m RNA were related to tumor differentiation degree, lymph node metastasis and TNM stage(P<0.05),but not related to the age, gender and tumor locations(P>0.05). The positive rates of ABCG2 m RNA were 36.7%(11/30),10%(5/50),72.4%(84/116)in incised margin normal mucosa, colorectal adenoma and colorectal adenocarcinoma. the expression of ABCG2 among groups had statistical difference(P<0.05). The expression of ABCG2 m RNA were related to tumor differentiation degree, lymph node metastasis and TNM stage(P<0.05), but not related to the age, gender and tumor locations(P>0.05). Spearman showed positive relative among expression of MDR1, MRP1, ABCG2 m RNA in colorectal adenocarcinoma(P<0.05).These results suggest that: most of colorectal adenocarcinoma have MDR1, MRPl,ABCG2 mediated primary multidrug resistance,and the high expression of MDR1, MRPl and ABCG2 were closely related to the lymph node metastasis, TNM stage and tumor differentiation degree of colorectal adenocarcinoma. Meanwhile, there may be a synergistic effect among the three genes in the malignant biological behaviors of colorectal adenocarcinoma. Upregulation MRPl expression and downregulation of MDR1,ABCG2 in adenoma stage maybe related to the formation of colorectal adenocarcinoma. Combined detection the expression of MDR1, MRP1 and ABCG2 in colorectal carcinoma, helpful to evaluate the drug resistance status and the potential metastasis of colorectal adenocarcinoma, it may become a clinical chemotherapy and prognosis valuable reference index...
