| Objective:Three acupoints, “Quchi, Zhongwan, Fenglong ”, are selected as the prescription called Acupoints "Jiangzhi Decoction" to study the effect on hyperlipidemia rats in terms of changes in electric acupuncture frequency(2HZ,50 HZ,100HZ), duration(10min,20 min,30min)and frequency(1times/d,1times/2d,5times/6d)by orthogonal design and to select the best stimulation parameters; to observe the effect of electric acupuncture on adiponectin(ADPN)and hypersensitive c-reactive protein(Hs-CRP)of rats with dyslipidemia according to above results and to explore the possible mechanism of Acupuncture "Jiangzhi Decoction" in treating atherosclerosis(AS).Method:Part one: One-hundred and thirty male SD rats were randomly divided into 11 groups including a normal group(n=20), a model group(n=11). Treatment group are divided into 9subgroups:groupⅠ~Ⅸ(n=11). Normal groups are fed with basic feed everyday, and the rest of groups were fed with daily ration of high-fat diet, unlimited drinking water and continuously fed freely for 7 weeks. 3weeks later, 10 rats from normal group and 1 rats from the rest of groups are randomly selected and drawn blood from abdominal aortic for detection the contents of TG、TC、LDL-C and HDL-C, and the model was successfully established. Then rats from groupⅠ~Ⅸ are treated with acupuncture treatment based on L9(34) orthogonal table and samples are taken after 4 weeks to detect the contents of TG、TC、LDL-C and HDL-C of rats in each group and observe changes of liver by the light microscope.Part two: Thirty male SD rates were randomly divided into a blank control group, a model group and a “Jiangzhi Decoctionten” group, with 10 rats in each group.Rats in the blank control group were fed with basic feed, and those in the model group and “Jiangzhi Decoctionten” group are fed with high-fat diet. Rats in the control group were fed freely, rats in model group were not given acupuncture stimulation and rats in acupuncture group were operated base on the optimal stimulation parameters. Four weeks later, hypersensitive c-reactive protein(HS-CRP)and adiponectin(ADPN)in blood serum were examined by chemiluminescence.Results:1. At the end of three weeks of modeling, compared with normal group, contents of TC and LDL-C in rats serum in model group increased significantly(P < 0.05),and the content of HDL-C(P < 0.01) decreased, which shows that the model rats with dyslipidemia were successfully prepared. After the treatment, the contents of TC, TG and LDL-C in model group are still high(P < 0.01), and the content of HDL-C(P < 0.01) decreased, which indicates that the rats had been in a state of abnormal blood lipids during the experimental period.2. Intervention of electric acupuncture in arranging acupoints in formula can improve the fur color and feces of rats and other general conditions. The contents of TC、TG、HDL-C in serum of rats fed with high-fat diet decreased and LDL-C increased. One of the best stimulation parameters for rats TC is 100 HZ, 5 times /6 d, 10 min. One of the best stimulation parameters for rats TG of 50 HZ, 10 min,5 times / 6 d. The best stimulation parameters of rats HDL- C is 50 HZ, 1times / d,10 min. One of the best stimulation parameters for rats LDL- C is 100 HZ, 20 min,1 times/ d. The order of various factors(parameters) is: TG: rate, retaining needle time and frequency; HDL- C and TC: rate, frequency, the retaining needle time;LDL- C: frequency, rate, retaining needle time.3. Under the light microscope: livers of rats in the model group demonstrated a large number of gathered lipid droplets in the cytoplasm of hepatic cells, fat denaturalization, moderate to severe swelling of cells, disappearance of part of cell membrane, the membrane fusion, vacuole-shape of different sizes by HE staining, the damaged structure of normal hepatic lobule. Hepatic cells in the electric acupuncture group had been repaired at different levels, and no obvious difference in the extent of liver cells repair among electronic acupuncture groups.4. Compared with the normal group, the content of Hs-CRP of rats in the model group increased and ADPN decreased significantly(P<0.05). After the electric acupuncture treatment, compared with the model group, the content of Hs-CRP decreased(P<0.05)and ADPN increased significantly(P<0.05).Conclusion:1. Electric acupuncture "Jiangzhi Decoction" can improve fur and feces of rats and2. other general conditions, regulate the level of TG、TC、HDL-C and LDL-, repair hepatic cells. The combination of best stimulation parameters is: 100 HZ,10min,1times/d.3. The optimal stimulation parameters of electric acupuncture "Jiangzhi Decoction" can effectively decrease the level of Hs-CRP, improve the level of ADPN, so it can be drawn that Acupoints "Jiangzhi Decoction" can effectively regulate blood lipid abnormality, improve cardiovascular function and decrease the risk of atherosclerosis. |
