| Objective: The present study was to detect the expression of HPV infection in 940 cases of outpatient and different subtypes of HPV cervical lesions P16 protein to explore different HPV subtypes P16 protein expression differences associated cervical lesions.Methods: 940 cases of HPV geno typing were collected from July 2013 to July 2014 in the Department of gynecology and then analysis of their infection status.Selected highrisk HPV-positive and 218 patients with biopsy cervical biopsy results: Among 56 cases of cervicitis, CINI45 cases, CINII31 cases, CINIII36 cases and 50 cases of cervical cancer. And then follow the P16 protein assay kit(American Eddie Vence) steps on paraffin sections immunohistochemistry.Results:(1) in the patients, HPV infection rate reached 24.57%(231/940), the infection rate among all ages was statistically significant.A total of 15 kinds of high-risk subtypes(HPV16,52,58,53,33,31,18,51,39,68,56,45,35,59,66) and three low-risk subtypes(HPV6,11, 81), wherein the high-risk HPV16 subtypes in this region accounted for the highest proportion of 29.00%, followed by HPV52, HPV58, HPV53, HPV33, percentage,were 12.56%, 9.96%, 7.36%, 7.36%. The HPV18 type only 4.76%.(2) in all cervical lesions P16 expression differences were statistically significant(P<0.05). The urban patients with chronic cervicitis, CINI, CINII-III, HPVP16 positive expression rates of cervical cancer were 11.63%, 43.08%, 71.43%, 100%, with the severity of cervical lesions expression of P16 protein gradually enhanced expression of differences of P16 protein in cervical lesions was statistically significant(P <0.05).(3)HPV16, HPV16 non-high-risk subtypes expression of P16 protein were respectively83.58%(56/67), 21.85%(33/151), the difference was statistically significant(P <0.05).Conclusion:(1) Rizhao City cervical HPV infection with HPV16 infection rates highest,followed by subtype HPV52, HPV18 subtype but relatively rare, and most areas of our country to HPV16, 18 sub types mainly have some differences, this is the local district to develop HPV vaccine identified certain theoretical basis.(2) With the degree of cervical lesions is increasing, the rate of P16 protein expression was also higher and higher, the difference was statistically significant, indicating the degree of cervical injury more severe expression of P16 protein is also increasing.Pathogenesis of disease occurrence from the macro level P16 protein could be a betterinterpretation of the cervix, have improved the accuracy of cervical pathological grade,and thus more conducive to improve the consistency between the different diagnostic pathologist.(3) In the city of Rizhao the expression of HPV16 subtype P16 protein was significantly higher than non-high-risk subtypes HPV16, HPV16 infection may cause P16 gene mutation, resulting in suppression of P16 gene inactivation, causing cervical cells malignant value, and ultimately lead to cervical cancer. |
