Objective:Barrett's esophagus(BE) with intestinal metaplasia is regarded as the premalignant condition of esophageal adenocarcinoma. However,it is reported that the columnar-lined esophagus(CLE) with non-goblet cells has also the risk of malignant condition.According to recent literatures,SOX9,CDX2 and MUC2 are studied in Barrett's esophagus, BE with dysplasia(BD) and esophageal adenomcarcinoma and attempt to illustrate the origin of BE.The goal of this study is to compare the expressions of SOX9,CDX2,MUC2 in the BE,BD and CLE.Methods: The patients with identified in 24 cases of Barrett's esophagus(BE),6 cases of BE with dysplasia(BD), 30 cases of columnar-lined esophagus(CLE)with non-goblet cells and 30 cases of control. The expressions of SOX9,CDX2 and MUC2 were determined by immunohistochemistry. The integrated optical density(IOD)was evaluated by Image Plus Pro6.0 software.Results:(1)The MUC2 expressions between BE,BD,CLE and control groups were observed,MUC2 was obviously expressed in the BE group with BE vs.CLE(8165.75±7947.87 vs. 266.25±614.45,P<0.001),MUC2 was highly observed in the BD with CLE vs. BD(266.25±614.45 vs. 2093.04±2265.92,P<0.001).There was no significant difference between BE and BD group with the Bonferroni of P value(P>0.0083).(2)The expressions of SOX9 and CDX2 were detected between BE,BD,CLE and control groups,both of them were not significantly expressed in the BE,BD and CLE.(BEvs.CLE:18832±16769vs.14518±14454;BEvs.BD:18832±16769vs.6400±2772;CLEvs BD:14518±14454vs. 6400±2772,P>0.05).(3)In BE intestinal-type group, the expression of SOX9 was significantly higher thanCDX2 and MUC2(IOD 18832±16769.12 vs. 8496±11196,18832±16769.12 vs. 8165.75±7947.87 P<0.05). There no significant difference between CDX2 and MUC2.(4)The low and median dysplasia of BE with SOX9 expression was higher than CDX2 and MUC2(6400.28±2771.71 vs.918.43±1053.45,6400.28±2771.71 vs.2093.04±2265.92,P<0.05).There were no difference between CDX2 and MUC2 in BE and dysplasia.(5)In the columnar-lined esophagus group, the evidence of SOX9 showed more ascended than CDX2 and MUC2 respectively(14518±14453.55 vs.4977±7131.35,14518±14453.5vs266.25±614.45,P<0.05)the expression of CDX2 was more increased than MUC2 significantly(IOD 4977±7131.35 vs. 266.25±614.45 P<0.05).Conclusions:MUC2 is highly expressed in the BE and BD,it may become an important marker in the diagnosis of BE and BD. Compared with CDX2 and MUC2,SOX9 is obviously increased in the BE,BD and CLE. SOX9 may play a crucial role in the follow up of BE.
