Objective Our study was intended to extract and isolate the effect ingredient from Cyclocarya paliurus, and evaluate the hypoglycemic effect and mechanism in diabetic mice. This study may provide scientific basis for further development and application of Cyclocarya paliurus.Methods The composition of CP was extracted and isolated , and it was identified with the reaction named Molish and Fehling. The diabetic mice model ware established by alloxan injection, then randomly divided into five groups, three groups were orally supplied with the CP 300mg·k-1, 450mg·k-1 and 600mg·k-1 daily, the other two was administration Dimethyldiguanide (Dim) and the normal saline. After 15 days of treatment, we measure the blood glucose and weight. Then, these mice were executed and pancreatic tissues were paraffin embed, HE staining for observing islet cells morphology. The antioxidant activity of the CP and the mechanism of the hypoglycemic mechanism of this Chinese medicine herbs were investigated by testing tissue homogenate MDA, GSH and Fenton reaction. Results1,After extracting the polysaccharide from the leaf of Cyclocarya paliurus,we get the yellow solid. This sample is CP, which is identified by the reaction of Molish and Fehling. CP could decrease the serum glucose (P<0.05), maintain the weight and protection of the impaired pancreatic cells in DM mice. It doesn't affect blood glucose and body weight of normal mice.2,CP has strong antioxidant activity with concentration-dependent pattern. CP can release the effect of lipid per oxidation (LPO) on MDA generated spontaneously in liver of mice. The maximum inhibition rate is 40.4%. It also has obviously inhibitive effects on ant lipid per oxidation induced by CCl4, H2O2and Fe2+-VitC. The maximum inhibition rate are 47.14%,50.07%,41.01%. When the CP concentrations are less than 2 mg·ml-1, they show concentration- -dependent pattern. When the concentration extract 2 mg·ml-1, the law disappeared.Conclusions CP has the ability of lower the blood glucose and maintains the body weight of the alloxan diabetic. The effect of anti-lipid per oxidation, scavenging free radicals and protecting islets coudle is the hypoglycemic mechanisms of CP.
