Cyclocarya leaves are the dried leaves of Cyclocarya paliurus(Batal)Iljinsk,belonging to the genus Cyclocarya of the Juglandaceae.It has the functions of expelling wind and relieving itching,promoting body fluid and quenching thirst,lowering blood sugar,lowering blood pressure and lowering blood lipids.At present,research reports have isolated more than 100 kinds of compounds including flavonoids,triterpenes,organic acids and other types of compounds from P.In order to further study the pharmacological activity and pharmacodynamic material basis of P.chinensis regulating lipid metabolism,this paper took C.pennywort leaves as the research object,and systematically carried out the separation of chemical components in ethyl acetate parts of C.pennywort leaves and the regulation of lipid metabolism.preliminary pharmacodynamic experiments.In this paper,column chromatography and other separation methods were used to systematically separate the ethyl acetate extraction part of the ethanolic extract of A.The isolated compounds were identified and analyzed by means of one-dimensional and two-dimensional nuclear magnetic resonance spectroscopy,mass spectrometry and other techniques.In this experiment,a total of 15 compounds(1-15)were isolated from the leaves of A.chinensis,namely kaempferol(1),kaempferol 3-O-β-D-glucoside(2),kaempferol 3-O-α-L-rhamnoside(3),quercetin(4),protocatechuic acid(5),trans-phydroxycinnamic acid(6),Peucedanol(7),Coptichic aldehyde(8),Coumestrol(9),Methyl Chlorogenic Acid(10),Schisandrin(11),3-Caffeoylquinic acid ethyl ester(12),α-D-Glucopyranosido-β-D-fructofuranoside,Sucrose(13),Luteolin(14)and Kaempferol Phenol-3-O-α-L-arabinofuranoose(15).Including 6 flavonoids and their glycosides,3 organic acids,2 coumarins,1 phenylpropionic acid,1 polysaccharide,1alkaloid and 1 lignan Among them,7,8,9,and 11 were the first compounds isolated from the leaves of C.pennywort in this study,which enriched the compound library of C.pennywort.In order to explore the pharmacological effects of the leaves of the buckwheat in regulating non-alcoholic fatty liver disease,the anti-inflammatory activity of the extracts of the leaves of buckwheat and the isolated compounds were screened in this study.Firstly,an LPS-induced RAW264.7 macrophage inflammation model in mice was established,and the anti-inflammatory activities of the compounds and extracts from the leaves of A.chinensis were studied.The results showed that the extracts and the monomer compounds from the leaves of A.chinensis inhibited the formation of NO to varying degrees,and the flavonoids had a better inhibitory effect,among which the compounds with better inhibitory activities were kaempferol and lignans.The inhibition effect of these two compounds with better activity on the m RNA expression of inflammatory factors was verified by RT-PCR experiments.In this study,a non-alcoholic fatty liver model was constructed in C57BL/6J mice induced by methionine and choline deficient diet(MCD)to study the regulatory effect of C.chinensis on hepatic lipid metabolism.The results of the study showed that gavage administration of the ethanolic extract of the leaves of A.chinensis could effectively improve the liver injury caused by MCD,improve insulin resistance,lower blood sugar levels,and inhibit the accumulation of lipids in the liver.In addition,the results of PCR experiments showed that the activity of A.chinensis in regulating lipid metabolism was related to the PPARα signaling pathway.
