The present work aimed to separation and identification flavonoids from jujube at Ruoqiang in Xinjiang using macroporous resin, HSCCC, HPLC and LC-MS. We analysed the hypoglycemic activities and mechanism of flavonoids from jujube with non enzyme glycosylation experiments and type I diabetic mice model induced by STZ, which provide experimental basis to research and develop hypoglycemic health products of red jujube.The main results were as follows:1. With the extraction rate of flavonoids as the index, the optimal separation conditions with macroporous resin S-8 to separate flavonoids from jujube water extract(group A) were as follows: the concentration of sample solution is 45 mg/m L,the value of p H is 5, the adsorption time is 2.5 h, the concentration of ethanol elution is 50%, the elution volume is 2 BV, the flow rate is 1.5 BV/h. With this condition, the content of total flavonoids ketone was increased from 0.16% to 2.23%, the purification multiple is 13.9.2. The Purification and identification of flavonoids of ethyl acetate extracts from jujube(group E). Fraction I and fraction II was separated and purified from group E with the methods of HSCCC and HPLC, which were identificated to be rutin and Luteolin C-hexoside by HPLC-MS.3. The hypoglycemic activities in vitro of flavonoids from Chinese jujube were evaluated by non-enzymatic protein glycosylation.The results indicated that the highest inhibition rate of early non-enzymatic protein glycosylation was the does of0.01 mg/m L of group A(22.52%).The highest inhibition rate of advanced non-enzymatic protein glycosylation was the does of 10 mg/m L of group B(54.01%).And the dose of 10 mg/m L of purified fraction I and fractions II have more significant hypoglycemic effects compared to positive drug metformin and group E(P<0.01), it showed that the higher purity of flavonoids in red jujube were, the more significant hypoglycemic effects of them have.4. Evaluating hypoglycemic activities in vivo of flavonoids from Chinese jujube with the method of type I diabetic mice induced by STZ. The results showed that group A and group B were both increased the level of body weight and decreased blood glucose, improved the organ coefficient of liver, kidney and spleen, reduce the content of serum insulin, increased the activity of SOD and GSH-Px, decreased the content of MDA of diabetic mice. But there were no significant improvement in the four indexes of lipids of serum of the diabetic mice treated with group A and group B.Besides, group A and group B can reduce the change of pathology of pancreas, liver and kidney. The results showed that group A and group B have hypoglycemic effects in the type I diabetic mice.
