Objective: This research investigated the effects of Kuijiekang by different soding route on ulcerative colitis induced by OXZ in rat, through observing the eating, fur, color, activity, and body weight and growth of rats; stool; colon specimens; and other intestinal mucosal injury and repair, to evaluat the therapeutic effects of Kuijiekang for treatment of oxazolone-induced ulcerative colitis and to provide experimental evidence for clinic.Methods: 1. Establishing the rat model of ulcerative colitis induced by oxazolone. The 12 rats were randomly divided into model group and normal group(n=6), half male and half female. The general situation was observed, including rat eating, body weight, stool, fur color, activity and mental state, situation stool(fecal occult blood, bloody stools, diarrhea). One week later the mice were sacrificed, and then take the colon tissue, colon tissue damage in rats, confirming the formation of ulcerative colitis. 2. Grouping: 120 rats were randomly divided into 12 groups. Normal control group, model group, Kuijiekang high dose group, middle dose group, low dose group, positive control group(n = 10), half male and half female. 3. Medical treatement: First day drop oxazolone sensitized rats’ back, 24 h later start given different concentrations of test drug intervention, once a day, followed by a continuous line 4d oxazolone enema modeling. Keep continuous drug intervention 6d. The observation continued until 24 h after the last administration. 4. Efficacy evaluation. Observe the general situation, including rat eating, body weight, stool, fur color, activity and mental state; situation stool(fecal occult blood, bloody stools, diarrhea), in DAI score every day. After the end of the experiment the results recorded including Kuijiekang enema administration or oral administration on rat spleen and thymus index, serum interferon-γ(IFN-γ), intestinal mucosa glutamine(Gln), interleukin-l(IL-1), interleukin-4(IL-4), and interleukin-10(IL-10) influence and the effect of drugs on colon tissue injuries ofscores and pathology.Results: 1. OXZ-induced model of ulcerative colitis can cause weight loss in rats, to the naked eye or bloody stools positive for fecal occult blood, DAI score higher. After modeling experiments confirmed OXZ enema can induce the formation of ulcerative colitis. 2. The gavage treatment group, high-dose enema treatment groups and middle-dose enema treatment group DAI scores were significantly lower than the model group, with significant difference(P<0.01). Serum levels of IFN-γ and IL-1 display no significant difference between drug intervention group and model group. Compared with model group, drug intervention group can reduce the serum levels of IL-4 and IL-10, which positive control group, high dose group and middle dose group compared with the model group has significant difference. The positive control group and Kuijiekang gavage dosing group and can increase the serum levels of Gln, but has no significant difference. Drug intervention group can reduce Organization score of model group, especially in high dose group was most pronounced(P<0.01, P<0.01). 3. The therapeutic effect of Kuijiekang enema administered is better than oral administration.Conclusion: Kuijiekang is effective for oxazolone-induced Ulcerative Colitis in rats by both enema and gavage. This therapeutic effect is more obviously in Kuijiekang enema administered in high dose group. |
