Objective:To research expression of nuclear factor Kappa B(NF-?B),TRAIL and MGMT protein to MGMT protein expression and relationships among them in primary glioblastoma.Methods:specimens of 67 cases have been taken during surgical operations of gliomas in the Neurosurgery-Departments of Handan Central Hospital and The Second Hospital of Hebei Medical university from June 2008 to August 2011,all the specimens were confirmed pathologically as primary glioblastoma by at least two pathologists.Expressions of NF-?B,TRAIL,MGMT in primary glioblastoma of the specimens of 67 cases were detected immunohistochemically;MGMT gene promoter methylation were tested by Methylation specific PCR(MSP).The result of present research were analyzed statistically by SPSS17.0 Analysis Software.Result:The result of immunohistochemical test of present research of 67 cases with primary glioblastoma showed that the positive expression of NF-?B was determined in 62 cases at the rate of 93%,the positive expression of TRAIL was determined in 62 cases at the of 82%,the positive expression of MGMT was determined in 34 cases at the of 50.7%,Among the 34 cases with positive expression of MGMT there were 30 cases with positive expression of NF-?B simultaneously.3 of 5 cases who were negative for NF-?B were determined to be negative for MGMT.There was a statistically significant positive correlation between NF-?B and MGMT(P<0.05).Among the 34 cases with positive expression of MGMT there were 32 cases with positive expression of TRAIL simultaneously.10 of 12 cases who were negative for TRAIL were determined to be negative for MGMT.There was a statistically significant positive correlation between TRAIL and MGMT(P<0.05).All the 62 cases with positive expression of NF-?B were determined to be positive for TRAIL and all the 5 cases with negative expression of NF-?B were determined to be negative for TRAIL,which showed a statistically significant positive correlation between TRAIL and NF-?B(P<0.05).Results from MSP show that MGMT promoter methylation happened in 20 cases(29.8%),among whom 12 cases were negative for MGMT.Among 47 cases without MGMT promoter methylation,negative expression of MGMT was determined in 25.There was no statistically significant difference between them.Conclusions1 Extremely high expression of TRAIL and NF-?B were determined in the primary glioblastoma tissue and there was a statistically significant positive correlation between TRAIL and NF-?B,even there also was a statistically significant positive correlation between both of them and MGMT.2 NF-?B and TRAIL both correlated with MGMT,they could be used as a new biomarker of predicting the chemotherapic efficacy to GBM.3 The quantity of MGMT measured inmmuniochemically could not completely and exactly reflect the state of MGMT promoter methylation.
