ObjectiveTo investigate the expression and difference of IDH1,MGMT and p53 proteins in primary and secondary glioblastoma and to analyze the prognosis of glioblastoma.MethedA retrospective analysis of 123 cases of glioblastoma,statistical analysis of the expression of IDH1,MGMT gene and p53 protein and its clinical data,comparing their expression and differences in primary and secondary GBM;analysis Correlation between IDH1,MGMT,p53 protein expression and its impact on prognosis.ResultThe OS and PFS of the primary GBM group were smaller than those of the secondary GBM group,and the difference was statistically significant(P<0.05).There were significant differences in IDH1 mutation,methylation status of MGMT promoter and positive expression of p53 protein between primary and secondary GBM(P<0.05).The IDH1 mutation status in primary GBM was associated with MGMT methylation status(P<0.01).Cox multivariate correlation analysis showed that the tumor site,MGMT promoter methylation status,and IDH1 mutation status were independent factors affecting the prognosis of patients with PFS and OS(P<0.05).Kaplan-Meier survival analysis showed that IDH1 mutation,MGMT promoter methylation and non-functional GBM had longer PFS and OS.Conclusion1.Secondary GBM has longer PFS and OS than Primary GBM.2.The IDH1 mutation rate,MGMT promoter methylation rate and p53 protein expression rate of patients with secondary GBM were higher than the original GBM,and there were statistical differences,supporting the different molecular inheritance of primary and secondary GBM.Pathway.3.The IDH1 mutation status in primary GBM is associated with MGMT methylation status.4.Tumor site,MGMT promoter methylation status,IDH1 mutation status are independent influencing factors affecting patient prognosis PFS and OS.IDH1 mutations,MGMT promoter methylation,non-functional GBM patients have longer PFS and OS.The expression of p53 protein was not associated with the prognosis of GBM.
