The Influences Of Tortoise-shell Glue And Antler Glue For The Expressions Of JNK And P38 MAPK In Guinea Pig Osteoarthritis Chondrocytes

Objective:By observing the effects of medicated culture of tortoise shell glue and deer-horn glue on the proliferation of Guinea pig's osteoarthritis chondrocytes and the expressions of JNK and P38 MAPK,to further define the degradation mechanism of tortoise-shell glue and deer-horn glue to adjust Guinea pig's osteoarthritis chondrocytes,to provide a new basis to expound TCM prevention and treatment of osteoarthritis by intervening in expressions of JNK and P38 MAPK in articular cartilage cells.Methods:1.The experiment was divided into four groups(tortoise-shell glue group,deer-horn glue group,glucosamine hydrochloride group and control group,respectively,in terms of G group,L group,A group and K group),to prepare each of group containing medicine culture.2.3-month-old Guinea pigs' OA chondrocytes was cultured and identified,and establishing the external cultured system.3.Each containing medicine culture that was respectively signed in four groups to the influence of the proliferation of F2 generation OA chondrocytes of guinea pig by MTT colorimetric method.4.Observing the expressions of JNK and P38 MAPK from each group Guinea pigs' OA chondrocyte by Western-Blot.5.Observing the expressions of the genes of JNK and P38 MAPK from each group Guinea pigs' OA chondrocytes by Quantitative PCR.Results:1.Type ? collagen immunohistochemical staining and toluidine blue staining were positive,indicating succeed in obtaining and establishing guinea pig's chondrocytes in vitro culture system.2.MTT assay result:Tortoise shell glue,deer-horn glue,glucosamine hydrochloride containing medicine culture for guinea pig's OA chondrocytes were promoting proliferation,the effect of proliferation was G group>L group>A group>K group(P<0.05).3.Western-Blot method Results:Tortoise shell glue,deer-horn glue and glucosamine hydrochloride containing medicine culture could inhibit the expressions of JNK and P38 MAPK,the effect of inhibition was G group>L group>A group>K group,(P<0.05).4.Quantitative PCR Results:Tortoise shell glue,deer-horn glue and glucosamine hydrochloride containing medicine culture could reduce the expressions of JNK and P38 MAPK mRNA,the effect of reduction was G group>L group>A group>K group,(P<0.05).Conclusions:Tortoise-shell glue,deer-horn glue can reduce the expressions of JNK and P3 8 MAPK mRNA in guinea pig's OA chondrocytes,and reduce the expressions of protein of JNK and P38 MAPK,that reduced the degeneration of guinea pig's OA chondrocytes,and delayed the pathological process of guinea pig osteoarthritis.
