The Effect Of Tortoise Shell Glue, Deerhorn Glue Containing Serum On The Activity Of Guinea Pig Articular Chondrocytes

Objective:To clarify the role of tortoise shell glue, antler glue treatment of osteoarthritis by observing the effects of Guilu Erxian glue split in the shell glue, deerhorn glue containing serum on guinea pig articular chondrocytes proliferation and the expression of MEK1/2 and ERK1/2, to provide a new basis for nourishing liver and kidney to promote articular chondrocytes proliferation for the prevention and treatment of osteoarthritis.Methods:1. The 48 Wistar rats were randomly divided into four groups (tortoise-shell glue group, deerhorn glue group, glucosamine hydrochloride group and control group, respectively, in terms of Group G, L group, A group and K group), respectively gavage for 7 days, blood samples were prepared containing serum.2.3-month-old Guinea pigs’articular chondrocytes cultured and identified, and establishing the vitro cultured system.3.Observing the effect of 5% containing serum for F2 generation chondrocytes proliferation by MTT colorimetric.4. Observing the expression of MEK1/2 and ERK1/2 from Guinea pigs’articular chondrocytes by ELISA.5. Observing the expression of MEK1/2 and ERK1/2 from Guinea pigs’articular chondrocytes by Quantitative PCR.ResuLts:1. Type Ⅱ collagen immunohistochemical staining and toluidine blue staining were positive, indicating succeed in establishing guinea pig chondrocytes in vitro culture system.2. MTT assay result:Tortoise shell glue, deerhorn glue, glucosamine hydrochloride containing serum on chondrocytes were promoting proliferation, G group>L group> A group > K group (P<0.05).3. ELISA method Results:Tortoise shell glue, deerhorn glue and glucosamine hydrochloride containing serum could promote the expression of MEK1/2 and ERK1/2, G group> L group> A group> K group, (P<0.05).4. Quantitative PCR Results:Tortoise shell glue, antler and glucosamine hydrochloride containing serum could significantly increase MEK1/2 and ERK1/2 mRNA expression, G group> L group> A group> K group, (P<0.05).Conclusions:Tortoise-shell glue and deerhorn glue can improve guinea pig articular chondrocytes expression levels of MEK1/2 and ERK1/2 and achieve the promotion of OA cartilage cell proliferation,and the role of Nourishig Kidney Yin-tortoise shell glue is stronger than Tonifying Kidney Yang’ deerhorn glue.
