In this paper, Viola yedoensis Makino.,a very important heat-clearing and detoxification drug, is taken as the main object to determine the content of fla vonoids and trace elements,and analyze the existence form of trace elements in it,to synthesize and characterize the flavonoids complexes and screen their im munocompetence. It laid the foundation for future in-depth study on material b asis and mechanism of Viola.This thesis is divided into 3 parts. Firstly, determine the total content of f lavonoids in 10 batches of Viola from different areas by UV spectrometer. Tak e total content of flavonoid as index, the optimum extraction process is obtaine d by single factor and orthogonal test investigation, and verified. Secondly, IC P-MS was used to determine the trace elements content in 10 batches of Viola from different areas. By measuring the content of different forms of trace ele ments, the possible existence form is discussed. Finally, based on the result of morphological analysis, synthesize complexes that may exist in Viola. NMR, I R, UV and other means of structural identification were used to identify the st ructure of complexes, and the immunologic competence of complexes were scr eened by determine the weight of immune organs.The result of total content of flonoid showed that, Viola from Hebei angu o and Shanxi has the highest content of flavonoid, respectively 3.99% and 3.6 4%. Content of flavonoid in Viola from different areas are different, this differ ence may due to the unlike environment condition. The results of optimum pro cess verification are stable; indicate that the process is reliable. The result of t race elements content determination showed the order of trace elements content s trace elements in Viola yedoensis was Ca>Mn>Zn> Cu> Pb>Cr>As>Cd> Hg, it suggests that the nature of Viola yedoensis itself is the predominant factor of trace element enrichment. In decoction, the contents of soluble Mn, Zn, Cu are higher, and organic trace element account for a considerable proportion. By chemical synthesis method, Mn, Zn, Cu complexes of flavonoids were synthes ized and characterized, six structures were inferred. In vivo active screening, n aringenin-Zn and naringenin-Cu had significant immune enhancement effect. Th ese results provide the basis for material basic research in Viola, and lay the f oundation for exploring the active mechanism of Viola.
