| Three plants belonging to Cayratia Juss of Vitaceae,namely,Cayratia Japonica Gagnep,Cayratia trifolia Domin and Cayratia oligocarpa Gagnep,which were common in Enshi Prefecture,were selected as the test materials in this paper.The effective components of three kinds of materials were studied.The major conclusions were drawn as follows:1.Three kinds of Cayratia plant had the highest content of flavonoid,which was their major active ingredient and mainly distributed in leaf.From the points of view of biomass and active ingredient content,Cayratia oligocarpa Gagnep was the variety of greatest development significance among the Cayratia three plants in the test,with flavonoid being the active ingredient with the most promising development significance.2.The ethanol extract of Cayratia oligocarpa Gagnep were analyzed as the main component:Rutin accounted for 40.41%of ethanol extract,while chlorogenic acid held 17.23%and quercetin took up 7.14%.The chlorogenic acid was first discovered in Cayratia Juss.In this article is the first time of Cayratia oligocarpa Gagnep ethanol extract were analyzed.Rutin as the main flavonoids of Cayratia oligocarpa Gagnep.3.The optimal extraction process of flavonoid in Cayratia oligocarpa Gagnep optimized by univariate and orthogonal test was shown as follows:solid-liquid ratio of 1:40,extraction time of 100 min,extraction temperature of 80℃,and microwave-aided time of 3.0 min.Extraction temperature had the greatest effect on flavonoid yield,followed by microwave-aided time,while extraction time and solid-liquid ratio did not have distinct effect on flavonoid yield.Under the optimum conditions,the yield of flavonoids was 131.93mg/g.Flavonoid content in Cayratia oligocarpa Gagnep was higher from June to October and lower from January to May.4.Coarsely purified Cayratia oligocarpa Gagnep flavonoid could significantly enhance the anti-oxidative capacity of mouse:in short-time continuous gavage test,dosage of 0~150mg/kg showed a positive correlation trend with efficacy,and anti-oxidative effects increased accompanied by increase in dosage.However,efficacy decreased accompanied by increase in dosage after reaching the dosage of 150mg/kg.Dosage for short-time continuous administration of Cayratia oligocarpa Gagnep flavonoid should not exceed 150mg/kg.In the long-term gavage test,coarsely purified Cayratia oligocarpa Gagnep flavonoid had excellent anti-oxidative effects on rats as the dosage increased,and the dosage showed a positive correlation trend with efficacy,which was superior to the positive control group within certain concentration range.In long-term gavage test,low dosage and moderate dosage of coarsely purified Cayratia oligocarpa Gagnep flavonoid could improve anti-fatigue in rats,while high dosage groups showed significantly reduced anti-fatigue effects. |
