| Objective Explored the proangiogenetic effect of LongZhi decoction,and explored the mechanism of proangiogenetic based on proangiogenic factors hypoxia induced transcription factor 1 alpha (HIF-la) and angiopo-ietin-2 (Ang-2).Methods ①60 SPF male SD rats with weight ranges from 270g to 310g were randomly divided into normal saline group(model group), Buy-ang Huanwu Decoction group( control group) and LongZhi Decoction gro-up(treatment group )with table of random number, 20 in each.②Constructe d qi deficiency and blood stasis in rats with acute cerebral infarction(QD-BSACI) models by exhaustive swimming for 14days plus modified middle cerebral artery occlusion(MCAO) operation.③Evaluated the rats’ neurolo-gyical function defection in Zea-longa scored scale method, administered normal saline (0.09g/100g), Buyang Huanwu Decoction (1.28g/100g), Long Zhi Decoction (1.36g/100g) respectively after MCAO operation.④Observ-ed the changes of the general state and weight of the model rats during the experimental period ,recoded the Zea-longa score after operation.⑤Ex-ecuted 10 rats randomly in treating 7th day and 14th day from three grou-ps respectively, observing the changes of microvessel,CD34,HIF-la and Ang-2 in their ischemic hemisphere in immunohistochemical way.Results ①After exhaustive swimming,rats appeared characterizations of qi deficiency and blood stasis, which with less ate, slow weight gain,loose stool, poor mental state, dry hair, light color nail, dark tongue that with ecchymos elt. ②After MCAO operation, rats appeared characterizati-ons of neurologic impairment with contralateral hemiplegia 、ipsilateral Ho-rner sign.③In the 7th ,10th day of treating ,the weight of the treatment gr-oup was higher than the model one (P<0.05), and in the 14th day, the weight of the control and the treatment group were all higher than the model one and the treatment group was higher than the control one (P all<0.05) .④In the 7th, 10th , 14th day of treating ,the Zea-longa score of the control group and the treatment group were all lower than the model one (P all<0.05).⑤In the 7th,14th day of treating,the microvessel of th e control group and the treatment group were all more than that of the model,and the treatment group was higher than the control one.(P all<0.05) .In the 7th day of treating ,the CD34 of the treatment group was highe r than the model and the control group (P all<0.05) ,In the 14th day of treating ,the CD34 of control group and treatment group was higher than the model one, and the treatment group was higher than the control one(P all<0.05), The CD34 of the 14th day of the model and the treatment group were significantly raise (P all<0.05) .The Ang-2 of the 7th day of the treatment group was higher than the model and the control group, the Ang-2 of the 14th day of the control and the treatment group were all higher than the model one, and the treatment group was higher than the control group (P all<0.05), the Ang-2 of the model group and the treat ment group were all raise significantly (P all<0.05) .The HIF-1α of the 7th, 14th day of the control and the treatment group were all higher than the model one, and the treatment group was higher than the control gro up (P all<0.05), in the 14th day of treating the HIF-1α of the tree group s were all raise significantly (P all<0.05).Conclusion ①The QDBSACI model which constructed by exhaustive swimming for 14days plus modified MCAO operation was tally with the clinical manifestation of QDBSACI.②LongZhi decoction can improve the general state and promote neural function recovery and angiogenesis of the ischemic brain tissue of rats with QDBSACI, the mechanism may be related to upregulate proangiogenetic factors of HIF-1α and Ang-2. |
