Objective:To investigate the effect and mechanism of methamphetamine on testosterone in male rats.Through the detection of testosterone and sex hormone levels,to observe the changes of testicular histology,analysis of the changes of 8-expression in the testis of hydroxy deoxyguanosine,to provide scientific evidence for the effect of MA on the level of testosterone in men and its related mechanisms.Methods:50 male SD rats were randomly divided into experimental group(n =230)and control group(n = 25).The experimental group and the control group were divided into 2 groups(n = 5)for each of the experimental group and the control group(n = 25)for a period of 3 days,7 days,14 days,28 days and 56 days.The experimental model was made by intraperitoneal injection of MA(8m/kg).On the 3 day,the 7 day,the 14 day,the 28 day and the 56 day,the rats were sacrificed by abdominal aorta,using light microscope to observe the morphological changes of rat testicular germ cell,using chemiluminescence method to measure the levels of luteinizing hormone and follicle stimulating hormone,serum and testicular testosterone,using enzyme-linked immunosorbent assay(ELISA)to determine the content of 8-hydroxy deoxy guanosine in the testis.Results:(1)The levels of spermatogenic epithelium were reduced in group MA rats,with only 2-4 layers where the cells arranged loosely,part of disorder,interstitial edema,part of spermatogonial cells and spermatocytes visible nucleus vacuolation and edema,necrosis of individual sperm cells.(2)Compared with the control group,3 days after the injection of methamphetamine,serum LH increased,but no significant difference(P>0.05),then the LH level decreased gradually,LH levels in 14 days decreased there were statistically significant differences(p<0.05),and with the extension time of MA injection decreased(28 days,56 days p<0.01).Compared with the control group,the serum FSH did not change significantly(P>0.05)at the initial stage(3 days and 7 days),and then the level of FSH gradually decreased(p<0.05 or p<0.01).(3)the level of 8-OHdG in testis increased gradually with the time of MA administration,and there was statistical difference at the 14 day(p<0.05).(4)3 days after MA administration,The serum testosterone to drug group than in the control group increased slightly,then testosterone levels decreased gradually,in the 28 and 56 day treatment group was statistically significant compared with the control group(p<0.05,p<0.01),and in 3 days in testis homogenate testosterone also increased,but no significant difference(P>0.05),then t the level of a downward trend,the testis homogenate in 14 days than the first serum testosterone was decreased(p<0.05).Conclusion(s):1?MA can cause pathological damage in male rats.2?MA increases the oxidative damage of DNA in the testis,and long-term use of MA may be a factor leading to oxidative damage in DNA,and it is time dependent.3?MA can also down regulate gonadal hormone LH,FSH,the serum testosterone and testosterone levels in rats decreased.The above results indicate that intraperitoneal injection of MA rats decreased testosterone secretion,the mechanism is through down-regulation of LH male rats,FHS increased oxidative stress in the testis,in both the secretion of testosterone in male rats was inhibited.
