[Objective]:To investigate the specificity and sensitivity of purified new bladder cancer(BC)monoclonal antibodies BCM29 and C3K5 in bladder cancer cell line,exfoliated cells in urine of patients with bladder cancer,bladder cancer tissues and circulating tumor cells(CTCs),make comparison with the conventional Papanicolaou stain method and preliminarily evaluate their value in the diagnosis and treatment of bladder urothelial carcinoma[Method]:Cell line of bladder cancer,tissue of bladder cancer,urine of bladder cancer and cancer cell of peripheral blood circulation of bladder cancer patient had been tested in this experiment.BCM29 and C3K5 antibody immunohistochemical method had been carried out directly upon the specimen of bladder cancer tissue;the specimens of cell line of bladder cancer,urine of bladder cancer and cancer cell of peripheral blood circulation of bladder cancer patient had been tested by employing the liquid-based cytology centrifuge.In detail,the specimen should be centrifuged on to the glass slide and then tests of Papanicolaou staining and immunohistochemical staining upon antibodies of BCM29 and C3K5 have to be carried out.Moreover,the specimens of urine and peripheral blood should be processed at first in order to remove the impurity or non-target cell,which should be centrifuged after the cell preservation liquid had been fixed.In a word,all of the tested specimens had to be observed under high-power microscope;BCM29 antibodies,C3K5 antibodies and Papanicolaou staining tests should be censused so as to figure out their sensibility and specificity upon the diagnosis of bladder urothelial carcinoma;potentials of clinical application upon the two kinds of antibody had to be evaluated preliminarily.[Results]:In this experiment,all specimens were treated with immunohistochemical staining,in which the cell line,urine and peripheral blood specimens were also tested with Papanicolaou staining.Under the microscope,it was found that both the two antibodies could bind to bladder cancer cells and the DAB staining results were positive.The specificity of BCM29 antibody against bladder cancer cells was up to 94.44%,and the sensitivity of C3K5 antibody was up to 76.92%.The staining results in EJ cells,T24 cells and bladder cancer cells were positive while the results in 293T cells were negative.Used commercial CD45 magnetic beads to separate circulating tumor cells(CTCs)from white blood cells(i.e.negative enrichment)and obtained a detection rate of 33%.[Conclusion]:It was found that BCM29 and C3K5 antibodies were two new monoclonal antibodies against bladder cancer.Based on the ThinPrep cytology test(TCT),using BCM29 and C3K5 antibodies to detect BC-related substances could be a rapid,noninvasive,low-cost and convenient method.BCM29 antibody showed excellent specificity and relatively high sensitivity;C3K5 antibody had good sensitivity but its specificity was relatively poor;both of them had better sensitivity and specificity compared with the traditional Papanicolaou stain.Therefore,this experiment preliminarily proved the clinical potential of this method and paved a way for further research on the mechanism of the two antibodies.
