The Preliminary Study On Theory Of Drug Property Of Typical Heat And Cold Medicine

Chinese herbal medicine is the core of Chinese medicine theory system.It is also prominent features of TCM.Chinese herbal medicine theory is summed up by long practices,and can be one of the basic principles of Chinese medicine clinical medication safety.It is of great significance in promoting the development of traditional Chinese medicine.Rhizom Coptis,Cortex Phellodendron,Radix Scutellaria is the cold medicine;Rhizom Zingiber,Cortex Cinnamomum,Fructus Euodia is the hot medicine.Zuojinwan,fanzuojinwan,ganlusan,zhuyuwan are the classic prescription which consists of Rhizom Coptis and Fructus Euodia.In this paper,the research on cold heat nature of traditional Chinese medicine can be divided two aspects:pharmacology study and metabonomics study.The effects of three kinds of cold medicine(Rhizom Coptis,Cortex Phellodendron,Radix Scutellaria)and three kinds of hot medicine(Rhizom Zingiber,Cortex Cinnamomum,Fructus Euodia)on the temperature,ATPase and SDH activity of the rats was studied in this paper.The activity of temperature,ATPase and SDH in cold medicine groups were decreased,but the activity of temperature,ATPase and SDH in hot medicine groups were increased.A metabolomic approach based on UPLC-QTOF/MS and integrative pattern recognition approach was developed for investigating the urine samples of rats.The long time has passed,the far the urine samples of model rats.from origi nal condition,and the samples of 21 day were the furthest.25 biomarkers con nected were confirmed and identified as L-2-Aminoadipic,acid,3’-UMP,(2R,2’S)-Isobuteine,(4R)-4-Hydroxy-L-glutamic acid,N-Acetyl-L-methionine,Malate,Citric acid,Melatonin,L-Phenylalanine,Tridecanedioic acid,9-chloro-10-hydrox y-hexadecanoie acid,5-Methylcytidine,Phenylacetylglycine,CDP-ethanolamine,3-Methylcytosine,p-Cresol glucuronide,Deoxycytidine,DL-Omithino-L-alanine,5-Hydroxyindoleacetic acid,5-Methyldeoxycytidine,5-Hydroxy-6-methoxyindolegl ucuronide,Formyl-5-hydroxykynurenamine,S-(Hydroxymethyl)glutathione,2,8-Di hydroxyquinoline-beta-D-glucuronide,(S)-N-(1,2-dicarboxyethyl)-Adenosine in col d medicine,and their interrelated metabolic pathway was aslo founded as Trypt ophan metabolism,Phenylalanine,tyrosine and tryptophan biosynthesis,Methane metabolism,Phenylalanine metabolism,Pentose and glucuronate interconversion s,Glyoxylate and dicarboxylate metabolism,Lysine degradation,Citrate cycle,Starch and sucrose metabolism,Glycerophospholipid metabolism,Pyrimidine me tabolism,Aminoacyl-tRNA biosynthesis.18 biomarkers connected were confirme d and identified as L-2-Aminoadipic acid,γ-Glutamyl-y-aminobutyraldehyde,1,4-Nonanediol diacetate,Malonic acid,DHAP(8:0),Histidinyl-Asparagine,L-gamm a-glutamyl-L-isoleucine,Threoninyl-Lysine,L-2-Amino-3-methylenehexanoic acid,L-allothreonine,N-Acryloylglycine,(S)-5-Amino-3-oxohexanoate,Ethyl trans-2-methyl-2-pentenoate,Gentisic acid,S-Glutaryldihydrolipoamide,(S)-1-Pyrroline-5-carboxylate,Cis-Aconitate,2-Oxoglutarate in hot medicine,and their interrelate d metabolic pathway was aslo founded as Citrate cycle,Lysine degradation,A anine,aspartate and glutamate metabolism,D-Glutamine and D-glutamate metab olism,Glyoxylate and dicarboxylate metabolism,Butanoate metabolism,Glycine,serine and threonine metabolism,Tyrosine metabolism,Arginine and proline m etabolism.Based on cold and hot medicine,the cold and heat property of zuojinwan,fanzuojinwan,ganlusan and zhuyuwan was studied.The activity of temperature,ATPase and SDH in Zuojinwan,ganlusan and zhuyuwan groups are decreased,but the activity of temperature,ATPase and SDH in fanzuojinwan group is increased.The long time has passed,the far the urine samples of model rats from original condition,and the samples of 21 day were the furthest.The result of PCA reveals,zuojinwan,ganlusan and zhuyuwan groups are partial to cold medicines,and fanzuojinwan group is partial to hot medicine.The above results suggested that the property of Zuojinwan,ganlusan and zhuyuwan are cold property,and the property of fanzuojinwan is hot property.In summary,the cold and heat property of the traditional Chinese medicine and the study of the scientific connotation of traditional Chinese Medicine was discussed and the theoretical guidance for clinical safe medication,experiment future research was provided in this paper,applying the study of relevant indicators and endogenous biomarker on energy metabolism of rats through modern advanced analysis methods,the theory of traditional Chinese medicine theory.
