| Objectives:In this study, the influencing factors of male infertility in Kunming were analyzed by analyzing the results of semen test in 110 cases. And the levels of seminal plasma protein in the two groups were detected by the proteomics in patients with oligoasthenozoospermia and healthy male to discuss the differences in protein between the two groups.Methods:The results were analyzed by computer aided semen analysis system (CASA) and analyzed statistically.Among them,we selected 10 male for childbearing age who meet the diagnostic criteria for oligoasthenospermia as a group,10 age-matched healthy male were recruited as a normal control group. Seminal plsma’s freeze-dried powder was collected in the both oligoasthenopermia group and normal control group. We applied Shotgun to detect the levels of proteins in oligoasthenospermia patients and health male. All test data was analyzed by the statistical software. P<0.05 was statistically significant.Results:1.110 cases of semen samples, semen volume <1.5ml of 16 cases, accounting for 14.55%; 1 case of pH <7.2, accounting for 0.91%;Sperm concentration less than 15×106 in 36 cases, accounting for 32.73%;The total number of spermatozoa less than 39 x 106 in 43 cases, accounting for 39.09%;Spermatozoa total activity (PR + NP) <40%of the 82 cases, accounting for 74.55%;There were 84 cases (76.36%) with forward movement (PR) less than 32%; Liquefaction time> 60min in 3 cases, accounting for 2.73%; Abnormal sperm> 96% of the 3 cases, accounting for 2.73%.2.There were 274 proteins in the patients with oligoasthenospermia compared with the healthy group (the reference level was more than three times), the difference was statistically significant (P <0.05).3.Among the 274 different proteins, 109 proteins were significantly higher than those in the healthy group, and the differences were statistically significant (P <0.05).Among them, 99 proteins were Health group was not expressed.4.Among the 274 different proteins, a total of 165 proteins were less than those in the healthy group, and were statistically significant (P <0.05). Among them, 144 proteins were oligoasthenospermia is not expressed in seminal plasma.5.We excluded the smaller protein fragments from unidentified proteins, in which heat shock proteins, keratin, epididymal secretory proteins, Ras proteins, histones, and prostatic stem cell antigens were more significant and discussed to demonstrate that they may all affect the occurrence of oligoasthenospermia and the development of the pathological process through different mechanisms .Conclusions: In the routine examination of semen, the decline of forward movement is the main factor affecting the semen quality of male in Kunming.The proteins in seminal plasma may be involved in the pathogenesis and development of oligoasthenospermia.They may be helpful to judge the severity of the disease, prognosis and efficacy. And they may influence each other in this process through synergistic effect. |
