| The direct complication of long time using antibiotic is alteration of intestinal flora,and furthermore result in antibiotic-associated diarrhea has been widespread phenomenon in clinic. Currently anthocyanins have been reported that it can affect the growth of the bacteria in vitro. But there is no report about the regulation function to disordered intestinal flora introduced by antibiotic in vivo.First antibiotic Cefixime Dispersible Tablets in mice, making the mouse intestinal dysbiosis model. Subsequent model mice were divided into Live Combined Bifidobacterium and Lactobacillus Tablets group, Blank control group, natural recovery group and anthocyanins from purple sweet potato group, were given the appropriate medication. Animals treated with appropriate medication for 14 days were euthanized by cervical dislocation; the ileocecum contents were collected in a sterile 2 mL centrifuge tube.The lysozyme solution was used for ileocecum contents total genomic DNA extraction.Analysis of the ileocecum contents microbiota structure by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) was carried out. Traditional culture results show: The model group compared with the blank control group, the Lactobacillus decreased,and the Escherichia coli,Enterococcus increased,the difference was very significant (P <0.05). PCR-DGGE results show: the diversity and richness of the bacterial community from the model group was lower than that of the normal blank control group’s sample. Antibiotic treatment resulted in reductions in the numbers of bacteria,Cefixime Dispersible Tablets of high and middle dose group can induce the disorder of intestinal flora in mice. The model was successfully established and the best choice was in the meddle dose.Ileocecum contents total genomic 16SrDNA genes were used to PCR-DGGE and quantitate intestinal bacteria by using real-time PCR(RT-PCR). PCR-DGGE results show:we have discovered that the diversity and richness of the bacterial community from Live Combined Bifidobacterium and Lactobacillus Tablets group, natural recovery group and anthocyanins from purple sweet potato group were lower than that of the normal Blank control group’s sample. The Live Combined Bifidobacterium and Lactobacillus Tablets group, anthocyanins from purple sweet potato group compared with natural recovery group,the difference of gray value was very significant(P<0.01). RT-PCR results show:Anthocyanins have a positive effects on the regulation of Bacteroides in intestinal flora.The Lactobacillus 16SrRNA gene copy numbers (per gram of wet weight of feces) were generally higher in the stool samples of anthocyanins from purple sweet potato group compared to the number in the stool samples of Negative control group. The Escherichia coli and Enterococcus 16SrRNA gene copy numbers (per gram of wet weight of feces)were generally lower in the stool samples of anthocyanins from purple sweet potato group compared to the number in the stool samples of Negative control group.Mice received by oral gavage antibiotic Cefixime Dispersible Tablets can be successfully produced mouse intestinal dysbiosis model. The APSP has a significant role in the adjustment of intestinal dysbiosis mice. |
