Objectives We observed the effect of acupuncture on the expression of Eotaxin,RANTES and TGF-β1 in the lung tissue of asthmatic rats in order to clarify the relationship between Eotaxin,RANTES,TGF-β1 and bronchial asthma and the effect of acupuncture on it.Methods 30 SPF male SD rats were randomly divided into 3 groups with 10 rats in each group.Groups were given interventions accordingly.The changes of pulmonary elasticity,lung compliance and inflammatory cells in alveolar lavage fluid in the rats were observed.The pathological changes of lung tissue were observed by HE staining.Enzyme-linked immunosorbent assay was used to measure the level of Eotaxin,RANTES and TGF-β1 in lung tissue,bronchoalveolar lavage fluid and serum.Light microscope was used to observe the expression of Eotaxin,RANTES and TGF-β1 in the lung tissue of asthmatic rats with immunohistochemistry and Immunofluorescence method.Results 1 In airway resistance: Compared with the blank group,the lung elastic resistance of asthmatic model group was significantly increased and the lung compliance significantly reduced(P<0.05).Compared with the asthmatic model group,the lung elastic resistance of the asthmatic model group was significantly lower than that of the acupuncture group,and the lung compliance increased(P<0.05),which was closed to blank group.2 In cell count: The number of white blood cells and eosinophils in the alveolar lavage fluid of asthmatic model group increased(P<0.05).Compared with the asthmatic model group,the number of white blood cells and eosinophils in the alveolar lavage fluid of the acupuncture group was decreased(P<0.05).3 The pathological changes in lung tissue were shown that,there were mucous plug formation in bronchial lumen of lung tissue in asthmatic model group,irregular bronchial epithelium,thickening of the airway smooth muscle,local rupture,luminal stenosis,and extensive inflammatory cell infiltration.The mucosal and erythrocytes of the lung tissue of the acupuncture group were significantly decreased,and the epithelial structure was more regular.The infiltration of inflammatory cells in the wall was also significantly reduced.4 The levels of Eotaxin,RANTES and TGF-β1 in the lung tissue,BALF and serum of the asthmatic model group were significantly higher than those in the blank group(P<0.05).Compared with the asthmatic model group,the levels of Eotaxin,RANTES and TGF-β1 in lung tissue,BALF and serum were significantly decreased in acupouncture group(P<0.05).5 The results of immunohistochemistry showed that the protein contents of Eotaxin,RANTES and TGF-β1 in the lung tissue of the asthmatic model group were significantly higher than those in the blank group(P<0.05).Compared with the asthmatic model group,Eotaxin,RANTES and TGF-β1 protein content decreased in acupouncture group(P<0.05).6 Compared with the blank group,the expression of Eotaxin,RANTES and TGF-β1 in lung tissue of asthmatic model group increased in the asthmatic model group(P<0.05).Compared with asthmatic model group,the positive expression of Eotaxin,RANTES and TGF-β1 was attenuated in acupouncture group(P<0.05).The overall observation showed,the expression of Eotaxin,RANTES and TGF-β1 in asthmatic model group was significantly increased,and decreased in acupounctrue group.Indicated that acupuncture can effectively control the expression of Eotaxin,RANTES and TGF-β1.Conclusions 1 The number of inflammatory cells in the lung tissue of asthmatic rats increased significantly,and it was involved in airway inflammation.Acupuncture treatment could significantly reduce the number of inflammatory cells to control airway inflammation.2 The expression of Eotaxin,RANTES and TGF-β1 in the lung tissue of asthmatic rats increased significantly,indicating that Eotaxin,RANTES and TGF-β1were involved in the pathogenesis of bronchial asthma.3 The contents of Eotaxin,RANTES and TGF-β1 in lung tissue of asthmatic rats were significantly reduced by acupuncture treatment,which indicated that acupuncture could decrease the expression of Eotaxin,RANTES and TGF-β1,and control the onset of bronchial asthma.4 The effect of acupuncture on bronchial asthma is closely related to Eotaxin,RANTES and TGF-β1.
