Ginseng and lycium barbarum play a key role in Chinese medicine as always.In order to further develop their medicinal applications,we use dermal fibroblasts and human B lymphoblasts to explore their skin-protective and immune activity.The focus of this study is whether ginseng effective component(Gx)can inhibit heavy ion irradiation and ROS-induced cell damage of fibroblasts and lymphoblasts.Additionally,a preliminary screening of the immunological activity of the active components of lycium barbarum was carried out.The results showed: 1)ROS inhibited the proliferation of primary human fibroblasts,induced abnormal arrangement of tubulin,integrin,and fibronectin,and inhibited the expression of MMP-1.Those effects could be alleviated by Gx.Gx protected ROS damaged dermal fibroblasts as follows: a)Improved the activity of damaged fibroblasts;b)Ameliorated fibroblast morphology;c)Restored the expression and arrangement of Tubulin,Integrin and Fibronectin;d)Inhibited the abnormal secretion of MMP-1 protein.2)Heavy ion irradiation down-regulated the proliferation of human B lymphoblast growth and induced cell apoptosis,ROS production and the abnormal expression of inflammation-related factors.Gx attenuated those situation.Gx played a protective role against injury of human B-lymphoblasts caused by heavy ion irradiation,as follows: a)Ameliorated cell morphology;b)Reduced cell apoptosis;c)Reduced intracellular ROS levels;d)Regulated the expression and secretion of TNF? and IL-1?.3)Five of 14 lycium barbarum components could increase the proliferation of oxidative stress-damaged B lymphoblasts and regulated the secretion of TNF? and IL-1?.They are 3 extracts of lycium barbarum wine(Gj1,Gj4,and Gj5)and 2 extracts of root bark(D2,D3).In conclusion,ginseng effective component(Gx)has a protective role on dermal fibroblasts and lymphoblasts.Moreover,the primary screening of active components in lycium barbarum showed 5extracts(Gj1,Gj4,Gj5,D2,D3)had immuno-biological activity.The results revealed that Gx and the effective components of lycium barbarum had potential value on development and utilization.
