Objective:To investigate the relationship between the serum level of sclerostin and valvular calcification in cllromc kidney disease pa Lients witn the CKD stage 3-5.To further explore the relationship between bone sclerosis protein and heart valve calcification in patients with CKD,and to provide a mechanism for the diagnosis and treatment of cardiovascular disease(CVD)in patients with chronic kidney disease.Methods:Collected 90 cases of CKD3-5 patients in our hospital,each group of 30 people.20 healthy volunteers(healthy control group)were selected from our hospital.Serum calcium(Ca),phosphate(P),alkaline phosphatase(AKP),HDL,LDL,TC,TG,blood urea nitrogen(BUN)and creatinine(Scr)were detected by Bayer ADVIA 1650 Beckman automatic biochemical analyzer in our biochemical room.C-reactive protein(CRP)was measured by Beckman Array 360 System.The serum and bone sclerotic protein levels were measured by enzyme-linked immunosorbent assay.Calcification of glomerular rate was calculated by MDRD simplified formula.By the heart of the hospital color chamber professional physician to measure heart valve calcification.Statistical analysis of CKD3-5 patients with osteosclerosis protein concentration and heart valve calcification and the relationship between the experimental indicators.Results:(1)Compared with the healthy control group,CKD3,4,5 patients with serum Ca decreased(P<0.05),PTH increased(P<0.05),CKD4,5 patients with blood Pi,calcium and phosphorus products increased(P<0.05);(2)The level of serum osteosclerosis protein increased gradually with the decrease of renal function from CKD3,and the level of serum osteosclerosis protein was significantly higher than that of healthy control group,and the difference was statistically significant(P<0.01);(3)Pearson correlation analysis showed that serum osteosclerosis protein was negatively correlated with glomerular filtration rate(e GFR)(r=-0.91,P<0.05)and serum calcium(r=-0.271,P<0.05),and positively correlated with serum creatinine(r=0.608,P<0.05),serum phosphorus level(r=0.295,P<0.05),PTH(r=0.334,P<0.05),calcium and phosphorus(r=0.275,P<0.05);(4)The proportion of heart valve calcification in CKD 5 patients was higher than that in healthy controls,the difference was statistically significant.(5)Logitic regression analysis of cardiac valve calcification showed that osteosclerosis was a risk factor for heart valve calcification in patients with CKD3-5.Conclusions:The concentration of serum osteosclerosis protein in CKD3-5 patients increased gradually with the decrease of renal function.Serum Sclcrostin was negatively correlated with glomerular filtration rate and serum calcium,and positively correlated with serum creatinine,serum phosphorus level,i PTH,calcium and phosphorus product.Serum Sclcrostin protein is an independent risk factor for heart valve calcification.
